Aluminum is the most abundant metal elements in the crust,which excessive intake and accumulation is associate with AD,Dementia Parkinson's syndrome and other diseases.Therefore,it is a research focus of discharge aluminum accumulated in the body.On the basis of preliminary experiment,we found that alumunim content in mice had decrease after lavaged jellyfish.In this paper,we further investigated the removing aluminum effects about treated jellyfish to healthy mice and enrichment of aluminum mice.Coupled with high-throughput sequencing technologies to explore the changes of intestinal flora in mice.And by using proteomics technology to study the roles of jellyfish row aluminum mechanism.As a kind of safe and effective new type of functional food to promote remove aluminum for jellyfish provide theoretical basis for new type of promoting the row of aluminum.The main research content is as follows:1?Study of salted jellyfish optimizing process method:In order to eliminate the influence of jellyfish itself aluminum content,in this paper,we studied these factors,as the ratio of material to solvent,immersion way,times of change water,immersion time for once,the kinds and concentrations of acid.All these factors would influence the removing salted jellyfish effort.The results show that under the way of stirring,using 1%acetic acid solution with solid-liquid ratio of 1:5 for 30 min and changing the water twice,the jellyfish aluminum residue(wet base)can be reduced to 15.497 mg/kg.The jellyfish through tissue homogenate for next experiments.2?The study of the removing aluminum effect of jellyfish to healthy miceThe healthy KM mice as the animal model,which respectively lavaged with normal saline(10 mL/(kg·d)),low(10 g/(kg·d)),medium(20 g/(kg·d))and high(30 g/(kg·d)))dose jellyfish.In order to research the influence of feeding jellyfish to the aluminum content of the different tissue and organs in mice.The results show that aluminum content had significant negative correlation with jellyfish lavaged dose.At the same time,we found that there is no obviously influence for lavaged jellyfish to other metallic element content in mice.These results preliminarily showed that lavaged jellyfish has a certain removing aluminum effect for healthy mice.3?The study of the removing aluminum effect of jellyfish to enrichment of aluminum miceLavaged healthy KM mice by AICl3 solution(80 mg/kg)for 7 days to build enrichment of aluminum mice model,then the aluminum content of different tissue and organs in these mice was determined.These mice was respectively lavaged with normal saline(as negative control),DMPS(as positive control,300 mg/kg),low,medium and high dose jellyfish(as experimental groups)for 14 days.The aluminum content of different tissue and organs in mice was determined and preliminary evaluation the removing aluminum effect of jellyfish to enrichment of aluminum mice.The results show that both lavaged high dose jellyfish and DMPS are highly significant reduce the aluminum content of different tissue and organs in mice,and it had no significant influence with other metallic elements content.These results showed that lavaged jellyfish has a certain removing aluminum effect for enrichment of aluminum mice.4?The influence of jellyfish to intestinal flora in miceStructure of intestinal flora take an important role in intestinal tract to change variety of nutrients for animals.We used depth of high-throughput sequencing technology to determine the intestinal flora of each experimental groups in Chapter 4,explored the influence of lavaged jellyfish to the structure of intestinal flora in mice,and studied the correlation between lavaged jellyfish and removing aluminum effects.The results show that the flora species in enrichment of aluminum mice significantly lower than healthy mice.No matter healthy mice or enrichment of aluminum mice,The abundance of intestinal flora diversity in mice were significantly increase after lavaged high dose jellyfish,especially the ratio of Lactobacillus increased significantly.All of these means that lavaged jellyfish had siginificant associations between aluminum content in mice and the structure of intestinal flora,and lavaged jellyfish would improve the intestinal health condition of enrichment of aluminum mice.5?The influence of jellyfish to the protein expression profiling of primary aluminum metabolism tissue and organ in enrichment of aluminum miceBlood and liver are the primary tissue and organ to accumulated and metabolic aluminum and other metallic elements in mice.In order to reveal the molecular action mechanisms of removing of jellyfish,we used the marked qualitative quantitative proteomic techniques to analyze the protein expression profiling of blood and liver in mice.The results show that lavaged jellyfish significantly changed the protein expression profiling of blood and liver,the differences induced protein are mainly involved in the process of metal ion transport,protein synthesis,oxidative stress,resistance to apoptosis,cell signal transduction and so on.During these processes,these protein family like Amine oxidase[flarin-containing]B,4-hydroxyphenylpyruvate dioxygenase-like protein,Metastasis-suppressor KiSS-1,Kininogen-1,and Ankyrin-3 are take an important role in the steady state of free metal ions intracellular in body,metal ion transport signal substance metabolism,signal transduction,the nervous system.Whese protein family are increase significantly.Which means high doses of jellyfish lavage may be induced by aluminum ions and transport related proteins,and the nervous system regulation of differentially expressed proteins,to promote removing aluminum,to maintain the body's physiological homeostasis.
