Toona sinensis leaves are rich in flavonoids,polyphenols and saponins substances,which have strong antioxidant activity.It is a good source of natural plant antioxidants.But,at present,the old leaves of Toona sinensis are often abandoned,not carried on the rational development and utilization,which resulted in the waste of resources and also is not conducive to cleaning up the environment.In this paper,using the abandoned old Chinese toon leaves as raw material,the edible alcohol as extracting agent,the effects of the different extraction methods were analysed and compared to get better methods of extraction and purification of antioxidants.Then the antioxidant extracts from Toona sinensis leaves with the chosen extraction and purification method were obtained.Antioxidant tablets were prepared by Proportional mixing the antioxidant extracts and other auxiliary materials.These results contribute to better use of the wasted Toona sinensis old leaves and improving added-value of Toona sinensis leaves resources..The main conclusions are as follows:(1)Ultrasonic-assisted extraction method can obtain higher antioxidant compounds and higher antioxidant activity than conventional solvent extraction and microwave-assisted extraction method.The three different extraction method have no obvious effect on the component of antioxidants from Toona sinensis leaf,and ultrasonic-assisted extraction method gained the higher contents of antioxidant monomers than the other two extraction methods.Results,by using response surface methodology coupled with a Box-Behnken design,indicated that optimal parameters for the ultrasonic extraction method was as the follows: Toona sinensis leaf size was 80 mesh,ethanol concentration 58.8%-60.6%(v/v),ultrasonic-assisted extraction time 40.7 min-41.3 min,solvent-to-material ratio 13.9-14.2m L/g,extraction temperature 60 ?,ultrasonic power 175 W and extracted for 3 times?.(2)The macroporous resin purification method was chosed for the better one,because of higher purification effect.The TFC and TPC of extract purified by macroporous resin are219.970mg/g and 521.033mg/g.By using the macroporous resin purification can obtain three and two more times of compounds than the unpurified extract and aqueous two-phase purified extract respectively.The optimum conditions of macroporous resin purification was as below:70m L extracts of 7mg/mL(including 3mol/L NaCl),flow rate of sample 2BV/h,80 mL 60%ethanol(pH 6)as eluent with the speed of 2BV/h.(3)The composition of antioxidative Toona sinensis leaf pieces were listed bellow :antioxidantive Toona sinensis leaf extract 3%,protein sugar 4%,dextrin 0.75%,magnesium stearat 13% and others as diluent(the proportion of starch / xylitol as 2).(4)Compared to steam sterilization or boiling sterilization,ultraviolet sterilization was perfect method for the pieces,The contens of total polyphenols and total flavonoids of antioxidative pieces from Toona sinensis leaf were 19.23 mg/ g and 8.34mg/ g respetively,and quercitrin was main individual phenol.
