Erinacines are a series compound with cyathane ditrepenes strutures,which mainly existed in mycelium of Hericium erinaceum.The biological test show the compounds have nerve growth factor-inducing activity,which have potential application in preventing the neurodegeneration including Alzheimer's disease.Although the compounds showed excellent biological active in in-vitro experiments,the low activity observed in in vivo tests,which may accounted by low solubility in water.Besides,the difficult of obtaining the componds also limited its studies.The thesis presented including four chapters.In Chapter 1,author firstly introduced the biological active compounds found in Hericium erinaceum,then described the biological actives and applications of erinacines.In second chapter,the separation of erinacine A were presented,and a HPLC analysis method were buildup for further works.Third chapter describe the works to optimize the extraction rate of erinacine A.Firstly,three kinds of mechanochemical extraction methods were tested showing the ultrasonic-assisted extraction was the optimal choice for the case.Then,an orthogonal experiment were performed to find the optimal condition for extraction.Besides,traditional extraction method were compared,while ultra-sonic extraction showed a much higher extraction rate.In the final chapter,the inclusion of erinacine to ?-cyclodextrin and 2-hydropropyl-?-cyclodextrin were studied.The work also used orthogonal experiment for optimization,and a fast stability test were performed.
