As the representative of anti-cancer drugs,paclitaxel is effective on most solid cancers,mainly existing in taxus species in nature.Because of the low content of paclitaxel in the yew,it is very necessary to determine a appropriate extraction process and to find a suitable separation and purification method.In this paper,the macroporous resin be use for purifying taxanes preliminarily,and the 3-aminopropyl triethoxysilane?APTES?as a functional reagent for preparation of modified mesoporous silica,will be separate and purify the paclitaxel from yew.The effects of adsorption performance relevent to paclitaxel were measured on HPD-400.et al eleven different types macroporous resin.It is found that HPD-400,HPD-100 and AB-8 adsorptive resins have a good effect for adsorbing taxtanes.HPD-400,HPD-100 and AB-8 were selected to study for the static and dynamic adsorption behavior in the actual taxane sample solution.And then purified by column chromatography which the select resins as carrier,the content of 10-DAB,cephalomannine and paclitaxel in loading sample will upgrade.In the artcle,using sol-gel method to prepare modified mesoporous silica with normal silicon dioxide as raw material,APTES as functional reagent.Setting paclitaxel adsorption capacity and selectivity as response value,Single factor method was use to optimize the preparation process.the added content of catalyst,Silicon source,functional reagent and porogen functions is investigated.Fianlly,the optimum conditions were obtained:catalyst hydrochloric acid?0.1mol/L?of 4ml,silicon source TEOS of 1ml,functional reagent APTES of 3.5ml,porogen F127 of 0.5g.Static equilibrium adsorption experiment of M-SiO2 and N-SiO2 for paclitaxel was studied.The pseudo-first-order,the pseudo-second-order models and Langmuir,Freundlich curve wrere used to evaluate the binding properties of modified mesoporous silica beads for paclitaxel.Combined with column chromatography technology,the separation and purification process of paclitaxel solution was conduct.Experimental results show that the modified mesoporous silica can be used for separation and purification of paclitaxel with higher recovery and purity.
