Abalone,one of the eight "Pearl",is rich in nutrition and has a very high medicinal value.The intestinal gland,gonads and other organs,occupied 1/3 of the abalone weight,were abandoned in the process.The phenomenon not only caused a great waste of resources,but also brought enormous pressure to the environment.Selenium polysaccharide is a kind of organic selenium compounds,the modified compounds kept polysaccharide original basic configuration and functions,and effectively improved the bioavailability of selenium and its physiological function.In this study,the abalone viscera were used as experimental material to extract abalone viscera crude polysaccharide(AVCP)by Ultrasonic combined with traditional enzyme.The optimum extraction parameters were studied.Through researched the process of sodium selenite modified abalone viscera polysaccharide,and obtained a new functional activity of selenium polysaccharide;finally,its antioxidant function was studied by conventional determination in vitro.Therefore,the research can provide experimental and theoretical basis for further study of abalone viscera selenium polysaccharide medicinal value.The study included:1?In the optimum conditions of extracting crude polysaccharide from abalone viscera,the ultrasonic power,enzyme concentration,extraction temperature,solid-liquid ratio,extraction time,the ultrasonic time effect were studied in the test by single factor and orthogonal experiments.The results showed that The optimum conditions for the extraction of polysaccharides were enzymatic hydrolysis time 1.5 h,Ultrasonic power 140 W and extraction time 45 min,the solid-liquid ratio 1:8,enzyme concentration 2.4×104 u·g-1 and temperature 45?.Under these conditions,the yield of was 7.75%.2?In the processes of selenium modified abalone viscera polysaccharide and structural characterization,the selenium content of abalone polysaccharide was used as response value,on the basis of single factor experiments,the best modification conditions were studied through the quadratic regression rotational combinational design.The structure of polysaccharide was characterized and analyzed by Fourier Infrared Spectrophotometry.The optimum conditions were the volume fraction of nitrate 0.6%,the selenium temperature 63.11? and selenium time 8.45 h,the addition of barium chloride 0.26 g,addition of sodium selenite 200mg.the selenium content of abalone polysaccharide was 564.08 ?g·g-1.The abalone viscera polysaccharide,abalone viscera selenium polysaccharide and sodium selenite were analyzed by infrared spectrum test,results show that abalone viscera polysaccharide had the SeO32-.3?The antioxidant activity of the abalone viscera selenium polysaccharide,abalone viscera pure polysaccharide and abalone viscera crud polysaccharide were studied by conventional methods in vitro.With the concentration increasing,the light absorption value improved,and the result showed that the abalone viscera selenium polysaccharide is better than other two kinds of polysaccharides;With the increase of concentration,three kinds of polysaccharides of superoxide anion radical(O2-·)scavenging ability increased in different degree;At the same concentration,the-OH scavenging ability difference,abalone viscera selenium polysaccharide-OH scavenging ability>abalone viscera pure polysaccharide>abalone viscera crud polysaccharide;Three kinds of polysaccharides of organic free radicals(DPPH)have good effect,when the concentration of 8 mg·ml-1,organic radical abalone viscera selenium polysaccharide,abalone viscera pure polysaccharide and abalone viscera crud polysaccharide(DPPH)scavenging rates were 77.5%,67.62%,68.18%.
