The abalone is a seafood with rich amino acid and low cholesterol and fat.It is well known as "the champion of seafood",and it has become the best-selling seafood in the domestic and international market.In recent years,with the increase of abalone production in our country,In the process of abalone,it mainly bases on primary processed muscles of abalone.However,lots of abalone viscera which account for 25%of the total weight have been abandoned,and only a small number of viscera are processed into fodder.This leads to the waste of resources and environmental pollution.Crude polysaccharide can be extracted from the abalone viscera to be polysaccharide health products,feed and spices etc.The added value of the abalone viscera can be increased,more economic and nutritional value can be created.Theoretical and technical support for the comprehensive utilization and development of abalone is provided.Researches on abalone polysaccharide characteristic index,the foundation for the deep processing and comprehensive development of the abalone viscera can be established,the basis for other related research can be provided.In this thesis,abalone viscera crude polysaccharide was extracted by pulsed electric field(PEF)&enzyme,then the polysaccharides were separated and purified and their characteristic indexes were studied.At first,the abalone viscera were handled.Then,abalone viscera crude polysaccharide(AVCP)were extracted by PEF&enzyme and 95%alcohol.Its extraction rate was 2.5%,its total sugar content was 61.62%and its protein content was 19.26%.Sevage method was used to remove its protein,the result showed that it could achieve the ideal effect by processing 4 times.The AVP which had been removed its protein was separated by DEAE-52,the neutral polysaccharide content was low,its yield was 6.53%.the acidic polysaccharides AVP-1,AVP-2,AVP-3's yields were 27.5%,43.7%,1 1.2%respectively.The chemical analysis showed that,AVP-1's total sugar content was 89.1%,its protein content was 3.99%and its sulfate content was 2.96%.AVP-2's total sugar content was 92.2%,its protein content was 2.13%and its sulfate content was 3.7%.AVP-3's total sugar content was 90.65%,its protein content was 3.68%and its sulfate content was 4.81%.Their molecular weights were determined by gel filtration chromatography,it showed that their molecular weights were 160694,78705 and 55081 respectively.IR analysis demonstrated that AVP-1 was a a-type glycosidic linkage,it contained pyranose and sulfate.AVP-2 was a-type glycosidic linkage,it contained pyranose and sulfate.AVP-3 was also a-type glycosidic linkage,it contained pyranose and sulfate.I determined their monosaccharide compositions by gas chromatography,the result showed that AVP-1 was composed of L-Rhamnose,D-Xylose,D-Mannose,D-Glucose,D-Galactose and Glucuronic acid;AVP-2 was composed of L-Rhamnose,D-Mannose,D-Glucose,D-Galactose and Glucuronic acid,and AVP-3 was composed of L-Rhamnose,D-Glucose,D-Mannose,D-Galactose and Glucuronic acid.
