people's requests for the beauty become increasingly high,the skin-whitening and lighting has become the pursuit of the majority of women,which makes whitening and removing freckle increasingly become the focus of the study.Now widely used whitening agent,such as kojic acid,vitamin C and its derivatives have poor stability,retarded effect and other shortcomings.Therefore,the exploitation of more safer and effective whitening products turning into an serious problem to be tackled.Because of Chinese herbal medicine being safer,mildness,less side effects and other characteristics compared to western medicine,so from the natural products to find safe and efficient skin-whitening active substances become more and more popular with whitening cosmetics research workers of all ages,showing a good development prospects.The herbs with the role of skin-whitening and freckle-moving is rich in resources in our country.Because of that,we have a unique advantage on study skin-whitening product.Now the Chinese herbal medicine used for the skin-whitening and freckle-removing products are mainly Angelica,Radix Paeoniae Alba,Magnolia,Salvia,licorice,ginseng and safflower and so on.In addition,the white poria is often used in many whitening products as a traditional whitening material,but there are little research and reports about its modern pharmacology on skin-whitening and how about their whitening effect.The researches and reports about Poria cocos can be used for skin-whitening and freckle-moving are few.Poria cocos is the dried sclerotium of Poria cocos(Schw.)Wolf.Its major chemical components are polysaccharides and triterpenoids.Some studies have shown that triterpenes have a good activities of anti-inflammation and anti-bacterial,freckle and warts removing,and modern pharmacology also initially proved that Triterpenes of Poria cocos have a variety of good biological activity of skin-whitening.Such as the effect of anti-bacterial,anti-inflammation,anti-oxidation,inhibition of tyrosinase activity and so on.Poricoic acid A as the main representative compound of 3,4-seco-lanosta-7,9(11)-diene type has the highest content in this type structure.And it has better water solubility with the structure of double carbonyl.While poricoic acid A used for whitening has not been reported.In order to further prove the skin-whitening effect of poria triterpene and to provide a theoretical basis and scientific basis of full used triterpenes of Poria cocos for the new application to skin beautifiness and skin care,we implement a lots of experiments in this paper to try to clarify the potential of Poria cocos in skin care.In this paper,triterpenes of Poria cocos were used as the main research object,and some experiments on mice B16 melanoma cells and antioxidant experiments and the experiments of inhibiting the catalytic activity of mushroom tyrosine and its mechanism of action were carried out.With that we can fully demonstration that triterpenes of Poria cocos would be used into whitening skin care products or not.The major research contents and results were shown below:1.Total triterpenes of poria have a certain antioxidant capacity and have a clearance effect of DPPH,ABTS free radicals,the IC50 value of clearing DPPH and ABTS free radical is 2892.19 ?g/ml and 180.75 ?g/ml respectively.2.Total triterpenes of poria and poricoic acid A had a significant inhibitory effect on the activity of diphenolase enzyme contained in mushroom tyrosinase and showed a dose-effect relationship.The inhibition effect of total triterpenes of poria was better than positive control drug arbutin in the range of 10.42-166.67 ?g/ml;poricoic acid A had the best inhibitory effect when its concentration reached 166.67 ?g/ml,the inhibitory effect is better than the positive control drug nicotinamide and arbutin.At the concentration of 200 ?g/ml,the total triterpenes of poria and poricoic acid A had a certain inhibitory effect on the activity monophenolase enzyme contained in mushroom tyrosinase,and at this concentration,the total triterpenes of poria,poricoic acid A and arbutin was not significantly different on inhibiting mono-phenolase activity.And they all were stronger than that of another positive control drug nicotinamide.They played a role mainly through combining with the dopaquinone formed by the oxidation of dopa and changing the hydrophobic environment of the amino acid residues contained in tyrosinase.3.Total triterpenes of poria and poricoic acid A have a certain cytotoxicity on the mouse melanoma cells.Maybe TTP and PA A can achieved the effect of whitening through inhibiting the proliferation of cells.The IC50 value of the inhibitory effect of total triterpenes of poria and poricoic acid A on cell proliferation was 118.26 ?g/ml and 48.01 ?g/ml respectively.And they had significant inhibitory effects on tyrosinase in mouse melanoma cells and showed a significant dose-effect relationship.When the sample concentration reached 12.5 ?g/ml,the inhibition rates were 40.86%and 39.53%respectively.Above all,by means of the experiments of antioxidant activity,inhibiting the catalytic activity of mushroom tyrosine and effecting generation of melanin,we provide a theoretical basis and scientific basis of full used triterpenes of the surface layer of Poria cocos for the new application to skin beautifiness and skin care.
