In recent years,with the rising prices of agricultural and sideline products and the pressure of environmental protection,the production cost of lincomycin has also increased year by year.Lincomycin as one of the company's products Topfond Pharmaceutical Co.,has an important position in the company.Therefore,how to improve the quality of products and reduce the cost of production to increase the competitiveness of the company has become an urgent problem.Topfond Pharmaceutical Co.Ltd.breeding a strain of lincomycin producing strain JS0214,a large number of tests carried out in the laboratory,the final titer should always put a bottle of 500?g/mL higher than that of the control strain(HPLC)above,but several test in the workshop in its potency has not produced bacteria high.Owing to the particularily of workshop production,we can not carry out a large number of tests,so our biotechnology research institute undertook the test.This study was to try to start the original production of bacteria as the foundation,through the observation of growth process of mutant dissolved oxygen,sugar,nitrogen,microbial concentration index,adjust medium proportion,optimizing control process,and ultimately improve the potency,reduce the cost,complete the test target.The main results are as follows:(1)Through the adjustment of grade one and grade two,and the fermentation medium of starch,glucose,soybean powder,corn sluny ratio,the indicators that can be reached by Streptomyces JS0214 forest forest production requirements.Lay the foundation for the smooth progress of the next experiment.(2)By adding KH2PO4 at different time in the middle and late stages of lincomycin fermentation,the cell senescence was delayed and the titer was improved.The forest can be thinner,fermentation in the process of the whole material,KH2PO4 together to re allocation for mixture ratio.In the simplified feeding process at the same time that Lin Kelian Aspergillus JS0214 draw-off titers reached 8200?g/mL(HPLC).By changing the fermentation process temperature,optimizing control process to size two in seed fermentation volume,making the forest Kelian Aspergillus JS0214 draw-off titers reached 8500?g/mL(HPLC),compared with the production of bacteria increased by 9%,to achieve the purpose of the experiment,and the pilot lay production base.(3)In the fermentation and feeding medium,gluten meal was used instead of a certain proportion of soybean cake powder to reduce the production cost,and the potency of the pot was also reached 8500?g/mL(HPLC).A new direction was found for the substitution of lincomycin for raw materials...
