In order to improve the edible value of Allium Victorialis and provide theoretical basis for further development and utilization of Allium Victorialis,the protective effect of Allium Victorialis extract on reproductive system injury in male rats was studied in this experiment.The water extract and ethanol extract of Allium Victorialis were obtained by reflux extraction.The model of deficiency of kidney yang was established by injecting hydrocortisone into male SD rats.The experiment was divided into blank group,kidney-yang deficiency model group,Jingui Shenqi pill positive control group,Allium Victorialis water extract and ethanol extract,middle and low dose group(1000 mg/kg,800 mg/kg,400 mg/kg),each group had 8 rats.From the 5th day after the model was made,the rats were administrated with the corresponding dose of Allium Victorialis extract and positive control drug every morning for 28 days.The changes of body weight,color and activity of the rats were observed during the course of modeling.Defecation and limb temperature were used to determine whether the model was successful or not.At the same time,the sexual behavior of male rats was observed,and the visceral coefficients of kidney,epididymis,testis,seminal vesicle and prostate were calculated after gastric perfusion.The contents of LuteinizingHormone(LH),Gonadotropin Releasing Hormone(GnRH),adrenocorticotropic hormone releasing hormone(CRH),testosterone,Malondialdehyde(MDA)and the activity of Superoxide Dismutase(SOD)were measured in serum.The morphological changes of testis were observed by H&E staining,sperm motility and motility in epididymis were analyzed,and monooxygenation in rat penis cavernous body was determined.Nitrided synthase(NOS)activity and cyclic guanosine monophosphate(cGMP)content.The results showed that the loss of appetite,drinking water and urine output increased and the activity decreased in the model rats.With the extension of the injection time,the body weight slowly increased,the body temperature decreased,the response was slow,the kyphosis in the back of the bow,sparse body hair,and loss of luster.Compared with the model group,the high dose group of the Allium Victorialis water extract significantly shortened the insertion latency,increased the insertion times and the number of riding span(P<0.05),but had no significant difference with the positive group.The results of the behavior were significantly shortened and the insertion and riding span increased significantly compared with the model group.The organ coefficient of reproductive organs in the treatment group was significantly higher than that in the model group,and the coefficients of kidney and epididymis had no significant difference with the positive group,and the testicular organ coefficient was significantly higher than that of the positive group.The organ coefficient of seminal vesicle prostate was significantly higher than that of positive group and blank group.The visceral coefficients of kidney and seminal vesicle in the ethanol extract group were significantly higher than those in the model group,but there was no significant difference between the positive group and the blank group.There was no significant difference in testicular organ coefficient between the high dose group and the positive group and the blank group,but the epididymal organ coefficient of the high dose group was significantly higher than that of the positive group.The serum testosterone content in the Allium Victorialis water extract group was significantly higher than that in the kidney yang deficiency model group(P<0.05).There was no significant difference between the middle and high dose groups and the positive group(P>0.05),but it was lower than that in the blank group.The serum testosterone content in the ethanol extract group was significantly higher than that in the model group,and the testosterone content in the high dose group was significantly higher than that in the positive group and blank group.The levels of A,A+B,and sperm viability were significantly increased in kidney-yang-deficiency rats than those in model group.The injury of testicular cells in kidney yang deficiency rats was improved after taking Allium Victorialis water extract.In the middle dose group,the testicular spermatogenic epithelial cells became rich,the tissues were arranged neatly,the seminiferous tubules were arranged neatly,the number of spermatozoa in the cavity became more,and the interstitial cells increased.The effect of the ethanol extract of Allium Victorialison the morphology of testis in rats with deficiency of kidney yang was similar to that of water extract,and the effect of high dose group on the repair of testicular tissue injury was better.The contents of CRH,GnRH and LH in serum of rats were significantly higher or lower than those of the model group.The CRH content in the high dose group was significantly higher than that in the positive group.The LH content in serum of rats with deficiency of kidney-yang was significantly higher than that of positive group and blank control group.The content of GnRH in serum of rats increased with the increase of water extract dose,and there was significant difference between different dose groups.The effect trend of ethanol extract of Allium Victorialis on sex hormone in serum of rats with deficiency of kidney yang was similar to that of water extract.Compared with the model group,the serum MDA content in the Allium Victorialis water extract group decreased significantly(P<0.05),and the MDA content in the high dose group was significantly different from that in the blank group(P>0.05).The content of MDA in the serum of rats treated with ethanol extract of Allium scallions was significantly lower than that of the model group.The content of MDA in the middle and low dose groups was basically the same as that in the positive group,but there was no significant difference between the two groups.The serum SOD activity of rats with kidney yang deficiency was significantly higher than that of the model group,and the SOD activity in the high dose group was significantly higher than that in the middle dose group and the low dose group(P<0.05).The serum SOD activity of the rats in the ethanol extract group was significantly higher than that in the model group,and the SOD activity in the high dose group was significantly higher than that in the middle and low dose group and the positive group(P<0.05).The activity of NOS and the content of cGMP in the corpus cavernosum of rats with deficiency of kidney yang were increased and improved after administration of Allium Victorialis extract.There was no significant difference in the activity of NOS between the high dose group of water extract and the high dose group of ethanol extract of Allium Victorialis.The cGMP content in the high dose group was significantly higher than that in the high dose group,but both were lower than those in the positive group.From the above results,it can be concluded that the extract of Allium Victorialis can improve the semen quality,promote sexual behavior,increase the weight of organs of reproductive organs and enhance the antioxidant function of the body in rats with deficiency of kidney yang.To increase the expression of NOS and the content of cGMP in the cavernous corpus cavernosum of the penis and induce the erection of testis to improve mating ability.Allium Victorialisis expected to be developed into functional food with positive effect.
