Lossing of freshness and producing of histamine often occured in fish and other aquatic products during storage,and it is affirmed that the ingestion of those aquatic foods may result in severe toxicological symptoms for human body.So it is significative for quality control of aquatic products to research the changes of freshness and inhibiting method of histamine during storage.In this dissertation,the simultaneous determination of biogenic amines in aquatic products by different derivatives and detection methods was investigated.Freshness indicators of blue scad(Decapterus maruadsi)including biogenic amines during storage were measured.The effect of histamine inhibitor was investigated.This dissertation is mainly consisted of three sections described as follows:Section one,the concentration of eight biogenic amines(BAs)by two different methods of high performance liquid chromatography(HPLC)were determined.For the pre-column derivatization,two different derivatization methods by o-phthalaldehyde(OPA)and dansyl chloride(DNS-Cl)were compared.The results showed that a linear response of BAs ranging from 1 to 100 mg/L could be achieved by two determining methods,the measurement precision(OPA method: 1.3~6.7%,DNS-Cl method: 2.6~9.9%,n = 6)were both less than 10%.But the measuring results of BAs in fish obtained by OPA method was significantly different(p<0.05)from those by DNS-Cl method.OPA method was simple,rapid and sensitive,and 3-fold of standard deviations of procedural blanks showed that the OPA method(0.03~0.35 mg/kg)was better than that(0.15~0.90 mg/kg)of DNS-Cl method;But the derivatives by OPA method was unstable,the detection spermidine and spermine was less sensitive.The DNS-Cl method was relatively complex,but it could be used to simultaneously determine all the eight BAs in fish including spermidine and spermine.In fact,we can choose corresponding derivative method based on the types and sample detection of biogenic amine.Section two,the concentration of biogenic amines?TVB-N value and total number of colonies during storage in the blue scad were determined.Results showed that the freezing(-20C)storage condition could inhibit the production of biogenic amines and TVB-N,while the concentration of biogenic amines and T-VBN values increased with storage time and storage temperature during the storage at 0C,4C and 25C.The concentration of cadaverine increased to 659 mg/kg at day 10 from the original value of 13.1 mg/kg,the concentration of putrescine increased to 103 mg/kg at day 10 from 16.5 mg/kg at day 6,TVB-N value also increased to 62.2 mg/kg at day 10 from the initial value of 24.9 mg/kg,in addition,the total number of colonies nearly doubl increased from during the storage at 4C.The concentration of histamine increased to 4720 mg/kg after being placed for 24 hours during the storage at 25 C.Therefore,the concentration of biogenic amine,TVB-N value and the microbial quantity could be quality indexes of fish during the storage.Section three,inhibition of histamine in blue scad by adding tea polyphenols,lysozyme,streptococcus and chitosan lactate were investigated,and appropriate inhibitors(carboxymethyl chitosan,tea polyphenols and lysozyme)were selected to investigate composite inhibitor for the production of histamine in fish during storage.According to the results of orthogonal experiment,the order of influence factors was: carboxymethyl chitosan>tea polyphenols>lysozyme,the qulity ratio of carboxymethyl chitosan,tea polyphenols and lysozyme of the composite inhibitor was 1:30:1,its adding amount in fish was 100 mL/kg,which could inhibit the production of histamine effectively.The inhibition rate of the composite inhibitor was above 90% after six days during the storage at 4C.Therefore,the composite inhibitor developed could inhibit histamine produced in fish during storage effectively.
