Confocal microscopy imaging technology has been widely concerned and researched by scholars because of high resolution,high precision and tomographic imaging capabilities.It has been developed into a confocal Raman spectroscopy system by combining the advantages of Raman spectroscopy with no damage,rich information,and no sample preparation.Therefore,laser confocal Raman spectroscopy has a wide range of applications in life sciences,chemical analysis,materials science,medical detection and other fields.In this article,a laser confocal Raman spectroscopy system has been designed based on confocal microscopy and Raman spectroscopy.At the same time,the microscopic topography information,three-dimensional map imaging and Raman spectroscopy information of the sample under test can be obtained.A method to improve the resolution of confocal Raman spectroscopy imaging has been researched.First of all,the development of laser confocal Raman spectroscopy system and the research status at home and abroad have been introduced.The detection principle of laser confocal Raman spectroscopy system has been introduced and analyzed.A laser confocal Raman spectroscopy system has been designed by combining simple confocal microscopy and Raman spectroscopy.Secondly,the various subsystems and their parameters of the laser confocal Raman spectroscopy system have been analyzed and discussed.The system experimental apparatus is analyzed and introduced.At the same time,the ZEMAX simulation analysis was performed on the optical path of the system,and the influence of the focusing and reflecting optical path of the microscope objective on the system was analyzed to determine that the overall optical path satisfies the size of the confocal pinhole and the analysis system has a good transmission efficiency,which is the structure detection system.The light path provides a theoretical basis.Again,the relationship between the focal length of the lens and the wavelength has been discussed,and a method for improving the resolution of the confocal Raman spectroscopic system has been proposed by adjusting the relative positions of the focusing lens and the confocal pinhole.The composition of the spectrum can be detected without changing the confocal pinhole size and ensuring the resolution.At the same time,the effect of confocal pinhole and lens aberration on the system has been analyzed theoretically.Finally,based to the theoretical analysis and the device system exploration,LED spectrum experiment has been designed to verify the feasibility of the proposed method,and the possible problems have been analyzed and summarized.According to the designed confocal Raman spectroscopy system,the experimental setup,improvement,debugging and analysis have been performed,the Raman spectroscopic information collection and result processing analysis of the tested sample has been performed using the experimental device of the laser confocal Raman spectroscopic system completed.The existing problems and possible causes have been analyzed and summarized,and methods for improvement have been proposed.
