The extraction and purification methods of polysaccharides were studied with red raspberry as the raw material,and their molecular weight and monosaccharide composition were determined and hypolipidemic effect was researched.The extraction process of polysaccharides was optimized by response surface methodology from red raspberry.The optimum conditions by compound enzyme method were optimized by response surface method.The results showed that enzyme(pectinase and cellulase)proportion,pH,material liquid ratio,temperature of enzymatic hydrolysis,enzyme hydrolysis time and enzyme addition is 1:2,4.0,1:30(g/mL),30?,25 min,1.5%(pectinase is 891U/g,cellulase is 1938U/g),respectively.The yield of red raspberry polysaccharides was 4.82%under these conditions.The polysaccharides of red raspberry were purified by DEAE-Sepharose Fast Flow ion exchange chromatography and Sephadex G-100 Sephadex column chromatography.Red raspberry crude polysaccharide was eluted with distilled water and different concentrations of NaCl solution.The flow rate was controlled at 1.33 mL/min.A single peak was recovered and named RRP.The RRP was further eluted by Sephadex G-100 Sephadex column chromatography to obtain a single symmetrical peak named RRP-IThe molecular weight and monosaccharide composition of RRP-I were determined by high performance liquid chromatography(HPGC)and gas chromatography(GC).The molecular weight of RRP-I was 11220.1845 Da,which is a homogeneous polysaccharide composed of glucoseHypolipidemic effect of RRP-I was according to animal experiments.The results showed that there were significant changes in the four indexes of TG,TC,LDL-C and HDL-C in the serum of rats with hyperlipemia after RRP-I was administered by 21 d.The serum levels of TG,TC and LDL-C in the low,middle and high dose groups of RRP-I showed a downward trend,while HDL-C showed an upward trend.These results show that RRP-I had the effect on hypolipidemic and have a certain dose effect relationship.
