No matter in war or peacetime,uncontrollable bleeding is the main cause for traumatic death,moreover,wound infection is also a great challenge in subsequent treatment.Due to the poor air permeability and tightness adhesion of wound,not only the existing hemostatic drugs or materials are inefficient,but the performance and efficacy of the materials are excessively simple.Therefore,it is of great practical significance to develop a high efficient hemostatic material which could be used in first aid,with good biodegradability,antimicrobial activity,and wound-healing effect.Chitosan(CTS)as a natural polysaccharide polymer can promote wound healing with preeminent antibacterial and hemostasis effect,has been widely used in the biomedicine field.In view of the dual challenges of uncontrollable bleeding and wound infection,chitosan was used as the raw material,which is composited with gallic acid and thrombin,and adopted the methods of chemical modification,physisorption,and UV cross-linking to develop a new chitosan-based composite hemostatic.And the hemostatic effect and antibacterial properties of this composite was evaluated and described as follow:1.Gallic acid was grafted onto chitosan thorough the chemical modification,synthesized and prepared the gallic acid-chitosan derivatives(GA-CTS).The inhibition zone method was carried out to evaluate the bactericidal effect on Escherichia coli.The observations supported that both CTS and GA-CTS have antibacterial properties.Besides the antimicrobial activity of GA-CTS is better than that of CTS.2.To modify the hemostatic properties of chitosan,Chitosan hemostatic microspheres were obtained by physically adsorbing thrombin drugs which is based on emulsification cross-linking method.Yunnan Baiyao was used as the reference drug,to evaluate the hemostatic properties and biocompatibility of microsphere,by measured with clotting time and mouse tail breaking test in vitro.It is indicated that the hemostatic effect of chitosan microspheres was significantly better than reference drug,and the microspheres was biodegradable.3.The chitosan-based composite hydrogel dressing was synthesized and obtained through acylation,UV spectrum cross-linking and physically mixing.Yunnan Baiyao was also used as the reference drug,and the antibacterial,hemostatic and wound healing abilities of the hydrogel dressings was studied by the inhibition zone test,mouse tail breaking test and mouse skin wound observation.It turns out that the hemostatic effect of chitosan-based composite hydrogel dressing was not only better than reference drug,but it displayed good role in biocompatibility and promote the healing of wound.To sum up,the chitosan-based composite hydrogel wound dressing is a promising hemostatic biomaterial which has very good performance in antibacterial,hemostatic,and wound-healing.
