Edible sources derived polysaccharide having a variety of medicinal value,is a kind of safe and non-toxic natural macromolecule.A large number of studies have shown polysaccharide from edible sources has various functions such as immuno-stimulating,antioxidation,hypoglycemic and hypolipidemic effects.Therefore,murine peritoneal macrophages were used as cell model in this study to evaluate the immunomodulatory activity of polysaccharides from seeds or leaves of Planago asiatica L.,okra and konjac.Then the immunomodulatory activity of screened polysaccharide was studied at whole,cell and molecular level in vitro and in vivo by the modern molecular biology method.The main research contents are summarized as follows:1.Preliminary screening of polysaccharides from edible sources on murine peritoneal macrophages RAW264.7.The effects of polysaccharides from seeds or leaves of Planago asiatica L,okra and konjac on the proliferation of murine peritoneal macrophages line(RAW264.7)were investigated.The results showed that PLWE-NAX(non-arabinoxylan polysaccharide from seeds of Planago asiatica L),PLWE-AX(arabinoxylan polysaccharide from the seeds of Planago asiatica L.water extract)had not obvious effect on proliferation of RAW264.7 in high concentration,and PLWE had no obvious concentration dependent effects.PLLCP(crude polysaccharides from Plantago asiatica L.leaves)and PLLP(sub-purified fraction)could enhance the proliferation in a dose-dependent manner.Meanwhile,the polysaccharides from okra peel named as OPWE-60,OPWE-100 and OPAE-60 had no significant effect on the proliferation.CWP(polysaccharide from Okra pulp)could promote proliferation,but also without any significant effect.For the konjac polysaccharide,KJ-22 was better than KJ-30 on the effect of proliferation,but weaker than the polysaccharide from Planago asiatica L leaves.In conclusion,PLLCP and PLLP had good effect on the proliferation of murine peritoneal macrophages RAW264.7 in a concentration-dependent manner.Therefore,polysaccharides from Plantago asiatica L.leaves were selected for further study on immune function.2.To assess the immunoregulatory activity in vitro of polysaccharides from the leaves of Planago asiatica L.,murine peritoneal macrophage cell RAW264.7 was used as cell model.The possible pathway was also explored.The results showed that PLLCP and PLLP significantly enhanced the proliferation,phagocytosis,NO release,TNF-? and IL-1? secretion of murine macrophages RAW264.7 in vitro,which was in a dose-depedent manner at the concentration from 20 to 160 ?g/m L.In addition,PLLCP and PLLP could affect the MAPK pathway,which was associated with murine peritoneal macrophages RAW264.7.3.To verify the immunoregulatory activity of polysaccharide from the leaves of Planago asiatica L.in vivo,immunosuppressive mice was subjected for the exploration.The results showed that PLLCP could significantly increase the thymus index,secretion level of immunoglobulin Ig A and Ig G in serum,activity of antioxidant enzymes such as SOD,CAT and GSH-Px in liver,and reduce the content of MDA.The results showed that PLLCP had immunomodulatory and antioxidant effects,which was consistent with the results in vitro.
