Pomegranate peel is a traditional Chinese herbal medicine with good antioxidant,antiviral,antitumor,antibiosis,immunity regulating activity due to polyphenols,polysaccharides,flavones and a variety of active ingredients in it.However,its effects were influenced by the low release of active ingredients and low bioavailability.In recent years,researchers at home and abroad mainly focused on the extraction process of active ingredients in pomegranate peel.However,the processing technology of pomegranate peel by using superfine grinding technology and physical-chemical property of superfine powder were not reported.Superfine grinding technology is a kind of emerging technology in recent years,and the key of this technology was to micrify the particle size,expand the specific surface area and help active ingredients get of the constraint of cell wall thereby improving the release of active ingredients,enhancing the bioavailablity and playing the efficacy of medicine.This experiment is based on pomegranate peel own characteristics,preparing pomegranate peel superfine powder by superfine grinding.These superfine powders make active ingredients release better,thereby increase the bioavailability in vivo,enhance the oral absorption and improve the efficacy of the drug.The main contents and results are as follows:1.The optimally grinding conditions for pomegranate peel were determined using response surface methodology(RSM).On the basis of single-factor tests,the effects of grinding time,grinding temperature,input quantity were explored.A regression model describing particle of pomegranate peel superfine powder as a function of three variables was established with the Design-Expert software according to the Box-Behnken design principle and experimental data.Quadratic regression analysis was used to establish a mathematical model describing particle of powder.The optimally grinding conditions for pomegranate peel were grinding time of 25 min,grinding temperature of-17?,input quantity of 198g(39.6%of feed ratio).Under the optimally grinding conditions,the minimum particle size of pomegranate peel superfine powder was 7.68 ?m,which is close to the predicted value of 7.96?m.Therefore,the established regression model has good prediction capability.2.Five types of pomegranate peel powders(A?E)were choosed by screening to investigate differences of physical-chemical properties.Superfine powder E shows the most superior physical-chemical properties in morphology,apparent density,angle of repose and slide,water holding ability and water solubility determination.Superfine powder E shows irregular state and uniform distribution by SEM determination.Apparent density could reach 0.751g/mL.The angle of repose and slide were 38.74° and 34.18°,respectively.Maximum water holding ability and water solubility index were 2.21 g/g and 56.5%,respectively.According to the FTIR and determination of total polyphenols,polysaccharides and flavones,the chemical construction of active ingredients were not changed after grinding.The release of polyphenols,polysaccharides and flavones were determined by simulating gastric fluid.Pomegranate peel superfine powder E has maximum active ingredient release in extraction of 60min.The release of polyphenols,polysaccharides and flavones were 15.87%,3.31%and 1.81%,respectively.3.We comparatively studied in vitro antioxidant activity of the pomegranate peel superfine powder E and coarse powder A by examining DPPH radical and ABTS+radical scavenging ability.The experiments show that,the radical scavenging ability of superfine powder E was better than the corase powder A.The scavenging value of DPPH radical and ABTS+radical could reach 98%and 72%,respectively.We comparatively studied the superfine powder group,coarse powder group,VE group and blank group in determination of vivo antioxidant activity.Experimental results show that superfine powder and VE could significantly improve the activity of SOD,CAT and GSH-PX and reduce content of MDA,compared with coarse powder and water in mice serum.The difference on vivo antioxidant activity between superfine powder and VE were not significant,but both of them were better than coarse powder.The experiment results indicated that the pomegranate peel superfine powder,which was processed through this experiment,is better than ordinary powder on particle distribution,physical-chemical properties,release of active ingredients and antioxidant activity.Thereby improve the bioavailability in vivo.
