1.Triazole benzamide derivatives are a class of potentially biologically active compounds.A synthetic route for the synthesis of triazolobenzylamine derivatives was explored.It is convenient to obtain 2-chloro-6-(1H-1,2,4-triazol-1-yl) benzamide using the 2,6-dichlorobenzonitrile as a raw material by triazole substitution,hydrolysis of nitrile,hydrolysis of amide,amidation of acid and other reactions as target compounds.2.The method of introducing 1,2,4-triazole into the 2-position of benzamide in the process was developed with the catalysis Cu I.The cross-coupling of 2,6-dichlorobenzonitrile and 1,2,4-triazole was fulfilled to get 2-chloro-6-(1H-1,2,4-triazol-1-yl)benzonitrile directly in the presence of Cu I and Cs2CO3.It provided technical support for the synthesis of the final target compounds.3.A method for the hydrolysis of 2-chloro-6-(1H-1,2,4-triazol-1-yl)benzonitrile to acid was established and it was found that 2,6-dichlorobenzoic acid could not be obtained smoothly by direct hydrolysis of 2,6-dichlorobenzonitrile in the presence of an acid or a base.So,the method of hydrolysis in two steps was determined.Firstly,2-chloro-6-(1H-1,2,4-triazol-1-yl)benzonitrile is hydrolyzed to the corresponding amide.The amide is then hydrolyzed to the corresponding acid by diazotization under the sulfuric acid and sodium nitrite.4.More than 10 2-chloro-6-(1H-1,2,4-triazol-1-yl)benzamide derivatives were synthesized using the obtained 2-chloro-6-(1H-1,2,4-triazol-1-yl)benzoic acid.Moreover,two 2-chloro-6-(5-thioxo-4,5-dihydro-1,2,4-triazol-1-yl)benzamide derivatives were synthesized.Intermediates and the target compounds were characterized and confirmed by NMR.
