Ginseng is a rare traditional Chinese medicine,the main substance containing ginseng saponin,sugars,amino acids,vitamins,proteins,peptides,organic acids,and rare ginsenosides are considered as the main active ingredient.They have the pharmaceutically active like anti-tumor,hypoglycemic,anti-oxidation and so on,especially the rare ginseng.But there is little or none rare ginsenosides in ginseng plants,so the effect on the activity of the rare ginsenoside and common research into rare ginsenosides exactly is the current focus of research scholars and priorities.The common method to transformation of ginseng saponins in vitro are the physical process,chemical hydrolysis,enzyme hydrolysis,microbial reforming process.The methods of physical process and chemical hydrolysis are simple but the specificity and efficiency are poor,Besides it can create constant pressure to the environment.The microbial reforming process has mild reaction condition,reaction selectivity and less by-products,simple post-processing and so on.But this method ask for the cultivation of microorganisms,need long experimental period and high operation requirements.However,enzyme method combines the advantages of the above methods in one,it is a good choice for transformation of ginseng saponins.The present study was prepared by an enzymatic method rare ginsenosides are more concentrated in selected corresponding enzyme from nature,the paper will be commodity enzyme preparation as a starting point to study the conversion of rare ginsenosides,ginsenoside obtained by the method of water extraction,quantitative analyzed by High performance liquid chromatography.10 kinds of glycosidases preparation(pectinase,cellulase,lactase([beta]-galactosidase),[beta] glucanase,cellobiase([beta]-glucosidase),[alpha]-amylase,?-amylase,hemicellulase,glucoamylase(amyloglucosidase,glucoamylase),glucose oxidase)and one protease preparations(trypsin)were studied the effect of ginseng root enzymatic conversion method with a view to screened high-efficiency conversion of ginsenosides merchandise enzyme preparation,here are the main findings:1?cellobiase([beta]-glucosidase),cellulase,hemicellulase enzyme has better effect.After dealing with ?-glucosidase enzyme,total saponin content of ginseng root 12.76% increase,ginsenoside Rb1 decreased 85.38%,ginsenosides Rh1 increased by 12.90%;total saponins after enzymatic hydrolysis of hemicellulose,ginseng root to improve 10.18%,78.21% increase ginsenosides Rg1,ginsenoside Rb1 reduced 42.11%,20.97% decrease ginsenosides Rh1;after cellulase hydrolysis,total saponin content of ginseng improved 8.72%,47.14% increase ginsenosides Rg1,ginsenosides Rb1 decreased 18.36%,17.72% ginsenosides Rh1 increase.In addition trypsin treatment can increase the content of ginseng root ginseng saponins.2?ginseng root extract with water ?-glucosidase enzyme,and the resulting rare ginseng saponin content is not digested sample comparative difference of about twice as much.Rd saponin content(3.56 mg / g)hydrolysis after about water extract(0.44 mg / g)10 times.Ginsenoside content increases because the role of ?-glucosidase enzyme,while the ?-glucosidase enzyme itself and the ginseng root itself,and independent of temperature.3?total saponins and rare saponins ?-glucosidase enzyme mixture with water extraction residue obtained are more than ?-glucosidase enzyme water extract of ginseng root,ginseng root so you can determine the fiber does not affect enzymatic effect.4?5 different companies ?-glucosidase enzyme hydrolysis enzyme optimization results show that the total ginsenosides obtained after the highest concentrations is: C's ?-glucosidase enzyme 72h(11.94 mg / g);give rare is the highest ginsenoside content: a company ?-glucosidase enzyme 12h(7.04 mg / g);Rb1 ? Rd highest conversion rates are: a company ?-glucosidase enzyme 12h(142.51%).5? Using the ?-glucosidase of Company A hydrolysis of ginseng root at different times(1-72h),the results showed that,A company ?-glucosidase enzyme ginseng root 12 h total saponins and rare highest saponin content,respectively,11.467.04 mg / g,7.04 mg / g.And Rb1 ? Rd conversion rate is relatively high,up to 142.51%.6?Before the enzymatic hydrolysis of insulation and rare ginseng root total saponin saponin transforming other effects,the results show that insulation helps improve total saponins,saponin content of rare and Rb1 ? Rd conversion.The total saponin content for 1h and then the resulting enzyme 24 h is 11.85 mg / g,a rare saponin content was 7.52 mg / g.The total saponin content directly 24 h enzyme obtained was 11.20 mg / g,a rare saponin content was 7.21 mg / g.
