Onion is a biennial herb of the genus Allium,which is rich in quercetin,prostaglandin A and other important biologically active substances,and becomes a vegetable with various nutritional values.Onion quercetin(OQE)is a polyhydroxy flavonoid extracted from onions.Modern researches show that onion quercetin has various effects,including anti-inflammatory,anti-oxidation,anti-cancer,blood sugar-lowering,immune regulation and cardiovascular protection.To investigate the anti-inflammatory effects of onion quercetin(OQE),1 ?g/mL lipopolysaccharide(LPS)was used to treat mouse peritoneal macrophages to establish a model of cellular inflammation,for researching the anti-inflammatory activity and mechanism of OQE in vitro.This provides an experimental basis for the development of anti-inflammatory functional foods.The research methods and results of this paper are as follows:1.In this study,a model of LPS-induced mouse peritoneal macrophage inflammation was established.The effects of OQE monotherapy group,OQE and LPS compound group on the phagocytosis of mouse peritoneal macrophages were detected by MTT assay;The results showed that 1 ?g/mL LPS treatment of mouse peritoneal macrophages for 24 h,In the dose range of 20~160 ?g/mL,OQE monotherapy group stimulation,OQE and LPS compound group all increased the phagocytic ability of mouse peritoneal macrophages.2.The effects of OQE on the secretion of LPS-induced mouse peritoneal macrophages cytokines and inflammatory mediators IL-1?,COX-2,IL-6 and TNF-? were detected by ELASA;The results showed that 1 ?g/mL LPS treatment of mouse peritoneal macrophages for 24 h,OQE treatment group can inhibit LPS-induced secretion of mouse peritoneal macrophages factors IL-1?,COX-2,IL-6 and TNF-?,and with the increased in OQE concentration,inhibition increased,there is a clear dose-effect relationship.3.The effects of OQE on the secretion of LPS-induced mouse peritoneal macrophages cytokines and inflammatory mediators gene IL-1?,COX-2,IL-6 and TNF-? mRNA expression were detected by Real-time PCR;The results showed that 1 ?g/mL LPS treatment of mouse peritoneal macrophages for 24 h,OQE treatment group can inhibit the mRNA expression of mouse peritoneal macrophages cytokines and inflammatory mediators gene IL-1?,COX-2,IL-6 and TNF-?,which were induced by LPS.With the increased in OQE concentration,inhibition increased,there is a clear dose-effect relationship;OQE also inhibits LPS-induced inflammatory response of mouse peritoneal macrophages and has anti-inflammatory effects.4.The effects of OQE in LPS-induced NF-?B and Akt mouse peritoneal macrophages signaling pathway protein phosphorylation expression levels were detected by Western blot;The results showed that 1 ?g/mL LPS treatment of mouse peritoneal macrophages for 24 h,LPS stimulation can significantly up-regulate NF-?B-p65 and Akt protein phosphorylation level in mouse peritoneal macrophages,while OQE-treated group shows opposite effects;Through the study of NF-?B and Akt signaling pathway,it was found that OQE can inhibit NF-?B and Akt signal transduction pathway and inhibit the expression of inflammatory factors,to exert an anti-inflammatory effect.
