Objective:The excellent strains of Marasmius epiphyllus were screened;the biological characteristics were investigated,and the optimal conditions for the growth of Marasmius epiphyllus slant were obtained.The deep fermentation process of Marasmius epiphyllus and the solid fermentation process were optimized to provide basis for industrial production;the fermentation product of Marasmius epiphyllus.The main components were determined to provide a reference for its quality control.The protective effect of Marasmius epiphyllus fermentation products on acute lung injury and the treatment of migraine were studied,and its mechanism of action and whether it replaced the existing drugs were explored.Methods:The excellent strains of Marasmius epiphyllus were screened and their biological characteristics were investigated by single factor investigation and orthogonal optimization experiments.The liquid fermentation process suitable for the growth of Marasmius epiphyllus was determined by changing the composition and culture conditions of the medium;The water content and the number of inoculation blocks were optimized to optimize the Marasmius epiphyllus solid fermentation process.The content of polysaccharides in the fermentation products of Marasmius epiphyllus was determined by phenol sulfuric acid method.The content of ergosterone and ergosterol in the fermentation products of Marasmius epiphyllus was determined by HPLC;Marasmius epiphyllus was obtained.The protective effect of fermentation products on acute lung injury in mice and the analgesic effect of nitroglycerin on migraine in rats were studied.The therapeutic effect of Marasmius epiphyllus fermentation products on the above diseases was studied.Results:The optimal conditions for the culture of Marasmius epiphyllus slant were: sucrose 1%,yeast powder 0.5%,MgSO4 0.05%,KH2PO4 0.1%,pH 5?6,temperature 25?30oC.The optimal conditions for deep submerged culture of Marasmius epiphyllus are: maltose 3%,yeast powder 2%,MgSO4 0.05%,KH2PO4 0.1%,pH between 5?6,and the best number of inoculation blocks is 12 pieces(100ml).For 140 ml,the number of days of canning is 10 days.The optimal water content of the Marasmius epiphyllus solid medium is 80% and the number of inoculated blocks is 6.The content of polysaccharide in the fermentation product of Marasmius epiphyllus was determined by phenol-sulfuric acid method:the content of polysaccharide in fermentation broth was 42.56mg/g,the content of polysaccharide in solid fermentation product was 33.43mg/g,and the content of polysaccharide in hyphae was 19.61mg/g.The content of ergosterol and ergosterone in the fermentation product was determined by HPLC: the content of ergosterol in the solid fermentation product was 0.0575mg/g,the content of ergosterone was 4.5?g/g,and the content of ergosterol in the hyphae was 0.13mg/g,the content of ergosterone is 1?g/g.LPS induced acute lung injury in mice.Marasmius epiphyllus fermentation products reduced the ratio of dry and wet weight(W/D)in the lungs and reduced the inflammatory response.In the nitroglycerin-induced migraine test,the fermentation product of Marasmius epiphyllus increased the levels of DA and NE in serum 5-HT and brainstem,and decreased the content of inflammatory factors.Conclusion:The excellent strains of Marasmius epiphyllus were screened to determine the optimal composition and culture conditions of the slant culture medium;the best liquid fermentation process for the growth of Marasmius epiphyllus was determined,the yield was improved;the best solid culture process was determined;and the ergosterol in the fermentation product of Marasmius epiphyllus was determined.,polysaccharide content and ergosterone content,provide a reference for its quality control;LPS induced acute lung injury in mice,Marasmius epiphyllus fermentation products reduce inflammation,regulate cell damage,indicating that there may be some treatment for acute lung injury Effect: In the experiment of nitroglycerin-induced migraine in rats,the fermentation product of Marasmius epiphyllus reduced the content of inflammatory factors in neurotransmitters,which may be the regulation of blood vessels,which may have some relief effects on migraine.
