With increasing of the consumption of crad,reports of allergy diseases induced by crab are growing,which has obtained widespread attention globally.Tropomyosin(TM)and Arginine kinase(AK)are panallergy present in crustaceans,which are important reason to induce food allergy.In the present study,we selected Scylla paramamosain as material.Firstly,TM was purified from muscle,and then the result show that Maillard reaction(wet,100°C,75 min,pH 8.5,TMrevertose weight ratio 1:3)with galactose could reduce its lgG binding activity effectively.The TM after Mallard reaction(TM-MR)digested by simulated gastric fluid(SGF)and simulated intestinal fluid(SIF)in vitro,the result of SDS-PAGE and immumoblotting indicated that the pepsin and trypsin hydrolysed TM-MR does not have lgG binding activity.The Circular Dichroism(CD)spectroscopy result suggested that helix conformation of TM-MR reduce 0.2% and ?-turn of TM-MR increase 0.9%.The phenol-sulfuric acid result showed that 0.33 mg galactose was expended by 1 mg TM after Maillard reaction.In a degranulation assay in RBL-2H3 cells,the ?-Hexosaminidase release rate was reduce from 58.33% to 21.37% by Maillard reaction.Secondly,AK was purified from muscle,and then the result show that Maillard reaction(wet,100°C,60 min,pH 8.5,AK-revertose weight ratio 1:3)with arabinose could reduce the lgG binding activity of AK effectively.The AK after Mallard reaction(AK-MR)digested by simulated gastric fluid(SGF)and simulated intestinal fluid(SIF)in vitro,the result of SDSPAGE and immumoblotting indicated that pepsin hydrolysed AK-MR hardly and trypsin hydrolysed AK-MR does not have lgG binding activity by 1 min.The CD spectroscopy result suggested that helix conformation of AK-MR reduce 32.5% and ?-turn of AK-MR increase 42.5%.The phenol-sulfuric acid result showed that 1.42 mg arabinose was expended by 1 mg AK after Maillard reaction.In a degranulation assay in RBL-2H3 cells,the ?-Hexosaminidase release rate was reduce from 54.88% to 20.37% by Maillard reaction.Finally,Maillard reaction(wet,100°C,60 min,pH 8.5,protein-revertose weight ratio 1:3,arabinose-galactosyl weight ratio 1:1)could reduce the lgG binding activity of crap muscle effectively which was analyzed by orthogonal array design.In conclusion,in the present study the optimal condition of Maillard reaction was confirmed to reduce the allergenicity of TM and AK.A hypo-Allergenic product about crab legs was contrived.
