In order to explore the effects of different tobacco leaves on the release of nicotine from snus,we built the simulated oral device and optimized the formula of artificial saliva in this study.Air or sun-cured tobacco samples were selected to study the influence of type,variety,pH and particle size on the nicotine release from snus,and the correlation between nicotine release and the main chemical components of tobacco leaves was investigated.The main experimental results are as follows:1.We built the oral device for simulating the environment of snus in oral cavity.The device consists of syringe,catheter,syringe jacket?2 mL?,water bath pot,test tube,automatic injection pump.Owing to the merits of easy disassembly and operation,the device could used for the treatment of samples in batches,greatly improving the efficiency in the research.2.In this study,the formula of artificial saliva was optimized through the comparison experiment and the orthogonal design experiment using artificial saliva and real saliva.The optimized formula was as follows:NaCl,0.4 g/L;KCl,0.4 g/L;CaCl2·2H2O,0.795 g/L;NaH2PO4·2H2O,0.78 g/L;Na2S·9H2O,0.005 g/L;Urea,1 g/L;carboxymethyl cellulose,3.0 g/L;pH,7.The artificial saliva could act as a alternative to the real saliva for the investigation of the nicotine release from snus in vitro.3.The nicotine releases from 58 types air or sun-cured tobacco samples were investigated.The results were as follows:When the nicotine content and pH were similar,the nicotine release from snus was related to the type of tobacco,among which the nicotine release from red sun-cured tobacco was relatively gentle,followed by rustica,and the nicotine release from cigar was the fastest.The release amount of nicotine from snus has nothing to do with the variety of air or sun-cured tobacco with the similar nicotine content and pH;The bigger the size of tobacco particles,the less the release amount of nicotine.As the extension of the release time,the release amount of nicotine with different sizes tends towards a convergence process.There was no significant relationship between the nicotine release rate and pH of the tobacco leaves;The cumulative nicotine release possessed extremely significant positive correlation with the content of nicotine,and significant positive correlation with volatile alkaloids content,total nitrogen content and chlorine content,while significant negative correlation with potassium content.4.The relationship between nicotine release and major chemical components was as follows:Early stage?0 min-6 min?:y=0.636xnicotine+0.6397,R2=0.7625;Medium term?6 min-15 min?:y=0.8143x nicotine-0.3214xpotassium+1.922,R2=0.9081;Late stage?15 min-30 min?:y=0.7834xnicotine+2.527xtotal volatile alkaloid-0.35,R2=0.9184.The content of nicotine and volatile alkaloid in tobacco leaves were the main influencing factors for nicotine release from snus.
