1,2,3-Triazole heterocyclic compounds have abundant physiological activities and have been widely used in medicine,pesticides,materials and other fields.Although researchers have done a lot of research on the preparation,structure and properties of these compounds,the development of efficient and rapid methods for the construction of 1,2,3-triazole structural units is still one of the hotspots in organic chemistry.Based on the advantages of organic synthesis techniques such as multi-component reactions and multi-component tandem reactions in the construction of heterocyclic compounds,several new synthesis methods of 1,2,3-triazole derivatives have been developed in this paper.The paper mainly includes the following three aspects:?1?A one-pot synthesis strategy for the synthesis of N,N-bis[?1,2,3-triazole-4-yl?methylene]tertiary amine derivatives has been developed.Reaction of aromatic primary amine and 3-bromopropyne in DMF-H2O mixed solvent in the presence of calcium hydride afforded N,N-di?prop-2-yn-1-yl?arylamine intermediate,followed by treatment with organic azide compounds catalyzed by Cu?OAc?2·H2O to form a series of newaromatic tertiary amines containing 1,2,3-triazole structure.This multi-component tandem reaction based on substitution reaction-[3+2]cycloaddition has the advantages of simple operation,high regioselectivity and good substrate adaptability,and the yield of the products is moderate to good.?2?A new method for the construction of 4H-1,2,3-triazole[5,1-c][1,4]oxazine-6?7H?-ones by three-component tandem reaction has been developed.The esterification of propargyl alcohol derivatives with bromoacetyl bromide was carried out in CH2Cl2 solvent promoted by pyridine.Then,the corresponding bromoacetate reacted with sodium azide under the catalysis of CuI to produce 4H-1,2,3-triazole[5,1-c][1,4]oxazine-6?7H?-one derivatives.The method has the advantages of simple operation,good compatibility of functional groups and good yields.?3?Hydroamination of 1H-1,2,3-benzotriazole with 1-allenyl-containing1,2,3-triazoles was investigated.Reaction of 1-allenyl-4-substituted-1,2,3-triazoles and 1H-1,2,3-benzotriazole in the presence of Pd?0?/dppp afforded a new class of allyl compounds containing 1H-1,2,3-triazole and 1H-or 2H-1,2,3-benzotriazole substituents.This new hydroamination reaction has good regioselectivity and high yield.
