The polysaccharide is a polysaccharide mixture extracted from the rhizome of the perennial herb of the genus Atractylodes,and the molecular weight of its polysaccharide components ranges from thousands of to tens of thousands of,which has the pharmacological effects of antioxidant,anti-tumor,anti-bacterial,immune enhancement and so on.At present,there are many researches on the extraction and purification process,molecular structure analysis and antioxidant study of polysaccharides,while the study on the chelation extraction process of polysaccharide and trace metal elements and the immunomodulatory activity of chelating objects has not been reported.Because metal ions have a variety of coordination and important catalysis in the organic body,when the metal ions and organic ligands form a stable complexes,the complexes can play a unique physiological and biochemical function;Therefore,the complexes formed by trace metal elements and polysaccharides not only have the biological activity of polysaccharide substances,but also can play the special function of trace elements.Therefore,the development of a new type of polysaccharide extraction and complexes preparation process can promote the biological activity of white polysaccharide and pharmacological value of a wide range of applications,has important practical value and scientific research value.In this paper,the new process of extracting polysaccharide from Radix albicans by hydrothermal alcohol sinking and metal chelation from the albino was explored,and the physical and chemical properties of polysaccharide products and animal immunomodulatory activity were studied by using trace element Zinc as raw material and micronutrient as chelating metal.The main research results are as follows:(1)Orthogonal test determined that the optimum technological conditions for the extraction of polysaccharides by hydrothermal zinc ion chelating are mixing proportion 0.8,pH value 5.0 and 1/10 of the material liquid ratio.(2)Ion chromatography and gel chromatography-differential-multi-angle laser light scattering analysis showed that the polysaccharides in the zinc complexes of the yellow-colored were mainly composed of glucose,galactose,arabinose,mannose and saline algae,and were small molecular polysaccharides with a heavy molecular weight of 2908,which was a high branching structure in the solution.The structure of high branching sugar chain was also verified by the analysis of polysaccharide bonding structure.(3)Through immunological experiments in SD rats,it was found that low doses of polysaccharide zinc complexes had significant immunomodulatory effect on immunosuppressive rats.
