A variety of dye molecules can form complex with optical properties by supramolecular forces with proteins.These complex will be used as a new type of protein-based fluorescent nanomaterials for biological imaging and drug delivery.In addition,some dye molecules have a certain fluorescence response to biological macromolecules,such as DNA,RNA,lysozyme,etc.Organelles play a vital role in living cells.Abnormal morphology and dynamics of cell compartments are early warning signals and hallmarks of a number of diseases.It is of significance to develop imaging tools for organelle monitoring,which is helpful in cell state determination and disease diagnosis.Ethyl Violet(EV),a representative of the triphenylmethane(TPM)dye family and heated bovine serum albumin(h-BSA),were used as building blocks to fabricate EV-BSA fluorescent protein nanovessels(FPNs).EV-BSA FPNs were as sembled likely through hydrophobic and electrostatic interactions between EV and h-BSA.EV-BSA FPNs are fluorescent-emissive with emission peak localized at 640 nm.Interestingly,EV-BSA FPNs can be readily internalized by mammalian cells and localized in both lysosomes and mitochondria.Moreover,EV-BSA FPNs can efficiently deliver irinotecan(CPT-11)into rectal cancer cells.The preparation method of such nanoparticles is simple,non-toxic and harmLess,with strong biocompatibility and excellent optical properties.We thus propose that EV-BSA FPNs serve as a dual organelle imaging tool and a carrier for drug delivery.Oil red O is a commonly used and so-called non-fluorescent dye for the tissue staining taking advantage of its capability in recognizing nitroglycerin and cholesterol esters.The supramolecular recognition between ORO and DNA was demonstrated by various means.After adding salmon sperm DNA,the emission peak at 600 nm increased obviously.The ORO fluorescence can be activated when mixed with DNA.The ORO dye showed high selectivity when test with a variety of biomolecules with similar structures or under biological background.Moreover,compared with other fluorescence DNA probes such as EB,ORO has the advantage of low cost and low toxicity.Therefore,ORO can act as a new double stranded DNA lighting up fluorescence probe.
