Effect Of White Lion Goose Serum On Gastric Cancer And Liver Cancer Cells

Goose blood is rich in nutrients and a variety of bioactive ingredients.It has been found that immunoglobulin,superoxide dismutase,thymosin,transferrin and other components in goose blood can not only enhance immunity and prevent cancer in humans,but also have the effect of antibacterial,anti-inflammatory,anti-oxidant and so on.However,due to the different breeds and feeding environments,the bioactive components and contents of goose blood are different.In this study,the excellent breed of goose-White Lion goose breeded by the Goose Institute of Heilongjiang Bayi Agricultural University was used as the research object to detect and analyze the active components and nutrients of the blood in it,and Then verified in vitro with gastric cancer and liver cancer cells to investigate the effect of White Lion goose serum on cancer cells.In order to compare the differences of bioactive components and nutrients in the blood of different breeds of geese under the same environment,White Lion goose,Haldor Baghi goose and Three flower goose 10 respectively were randomly selected to detect the trace amounts in blood with trace element analyzer;The amino acid composition of the blood is detected by an amino acid analyzer;the content of biochemical indicators in the serum is detected by an automatic biochemical analyzer;the level of immunoglobulin G(Ig G),superoxide dismutase(SOD),transferrin(TRF),thymosin(Thymosin)and glutathione(GSH)in serum are detected by ELISA.The results showed that magnesium,zinc and iron in the blood of the White Lion goose were significantly higher than those of the other two kinds of geese(P<0.05);Besides,the total serum proteinof the White Lion goose was significantly higher than that of the other two geese(P<0.05).Especially,the content of serum albumin is the highest.The White Lion goose contains 18 kinds of amino acids,the first and second limiting amino acids are tryptophan and isoleucine respectively,the level of total amino acid,essential amino acid,basic amino acid,lysine and the EAAI were higher than those of the other two kinds of geese;the level of serum Ig G,TRF and thymosin of White Lion goose were significantly higher than those of the other two geese(P<0.05),and the level of SOD in serum of White Lion goose was the highest.According to the above research results,the blood of three species of goose is rich in bioactive components and nutrients.The level of active components in the blood of White Lion goose is higher than that of the other two breeds.According to the functional active ingredients contained in the blood of the White Lion goose,the White Lion goose serum was extracted.Human gastric cancer cells(AGS)and human liver cancer cells(Hep G2)were used for verifing in vitro Cell proliferation was detected by CCK-8;Cell morphology was observed by inverted microscope;Wound healing ability of cells was detected by scratch and abrasion test;Cell cycle distribution and cell apoptosis were detected by flow cytometry.The results showed that with the increase of the activation concentration and the prolongation of the time,the serum of White Lion goose can reduce the vitality of gastric cancer cells(AGS)and liver cancer cells(Hep G2),change the morphology of gastric cancer cells and liver cancer cells,and reduce the cell migration rate.The proportion of cells in the early stage of DNA synthesis(G0/G1 phase)and late DNA synthesis(G2/M phase)of gastric cancer cells and liver cancer cells is decreased,the proportion of cells in DNA synthesis phase(S phase)is increased,and the apoptosis rate of cells is increased and decreased the number of living cells.It can be proved that the serum of White Lion goose has a significant inhibitory effect on the proliferation of human gastric cancer cells and liver cancer cells,and can change the distribution of cell cycle and induce apoptosis,and it is dose-time dependent.The results showed that the blood of the White Lion goose contained high bioactive components,which had obvious proliferation inhibition effect on gastric cancer and liver cancer cells cultured in vitro.
