In this paper,the biological activities and research progress of barbituric acid cyclic compounds were reviewed firstly.Based on the literature review,we investigated the difference in the reaction between barbiturate-based olefins and 1,3-dicarbonyl compounds to synthesize ring derivatives containing barbituric acid.The 1,3-dimethylbarbiturate-fused olefins were synthesized with 1,3-dimethylbarbituric acid and various substituted aldehydes taking place aldol condensation reaction.Reaction with different dicarbonyl compounds through the multistep cascade reaction gave the different products and the reaction conditions were optimized.A total of 84 new compounds were synthesized by this method,include 26 spirocyclopropane barbiturates,14 furo[2,3-d]pyrimidines and 44 spirodihydrofuran barbiturates.Their structures were confirmed by NMR,IR,high resolution mass spectrometry and single crystal diffraction.After that,we proposed a reaction mechanism for the multistep cascade reaction.
