Oat grain is one of the essential sources of water-soluble P-glucan polysaccharides.Linearly attached D-glucose monomers via ?-(1-4),?-(1-3)glycosidic linkages form the P-glucan polysaccharides.The ?-glucans exist together with other matrices of components,which imposes difficulties in the extraction and purification steps.Therefore,careftul attention must be given to efficiently isolate the purified form of the P-glucans to attain high level of physicochemical properties.Thus,we have successfully extracted ?-glucans from Oat grain using a simplified water soaking method and sodium phosphate buffer solution.Then the physicochemical results were compared with the control counterpart.Accordingly,the ?-glucans obtained with the soaking method could show the overall high quality over the P-glucans extracted using sodium phosphate buffer solution and control sample.Therefore,the purity of ?-glucans from the soaking method offered 91.27%,which is higher than that of the ?-glucans obtained from the sodium phosphate buffer solutions(75.45%)and control(66.68%)respectively.Regarding the physicochemical parameters(viscosity solubility,water binding capacity,protein and starch impurities),the soaking and sodium phosphate buffer methods both showed higher scores of(62.61 cP,56.25%,4.9 gg-1,1.01%and 1.70)for soaking,and(52.45 cP,64.02%,4.83 gg-1,2.63%and 1.77%)for the buffer solution than(49.56 cP,37.5%,3.11gg-1,3.02%and 3.27%)of the control methods respectively.This shows that the soaking strategy has higher efficiency compared with the sodium phosphate buffer solution and the control methods.Future work may be needed for further optimization of the soaking strategy to find high-quality ?-glucans.
