Trichoderma is a class of important saprophytic fung,which has strong adaptability to ambient temperature and pH,quick growth and reproduction speed,and excellent ecological competitiveness.Therefore,in many countries,it was registered as a core strain of biological insecticides,fungicides,microbial fertilizers,bioabsorbents and natural resistance stabilizers,and widely applied to agricultural production with its good effects of promoting growth,resisting diseases and increasing yield as well as has great potential for development and market space.Solid culture is the main way of producing most of Trichoderma.However,at present,the sterilization and disinfection of solid culture costs a lot and has a complex operation,which confines the scale solid production of Trichoderma viride.Therefore,it is very important to optimize the production condition of Trichoderma viride TV41 solid fungicide.In the article,amino acid washing wastewater generated by the enterprises of amino acid production was used to sterilize the solid culture medium of Trichoderma viride.Meanwhile,the conditions of Trichoderma fermentation including the amount of waste liquid and the time of adding,adding different mineral elements,inoculation methods and inoculation amount of seed liquid,different culture methods and so on were optimized.Finally,the effects of Trichoderma viride TV41 on tomato seedling growth under salt stress were studied.At first,the effects of different amino acid waste and the time of placement on the solid fermentation of Trichoderma viride TV41 in the condition of triangular flask-culturing were researched.The results indicated that the optimal amount of amino acid waste was 5 mL and the optimum time was 0 h per 10 g of bran and rice straw(5:5).Under this condition,the sporulation of Trichoderma viride TY41 reached 2.41×109 cfu/g.The optimum amount and time of addition of the amino acid waste were used to the following experimental study.Next,the effects of different seed broth,different inoculation amount and culture vessel on the fermentation of Trichoderma viride TV41 were studied.The results implied that when the mycelial of Trichoderma viride TV41 growing in oat liquid medium was transferred into solid fermentation,the best effect was seen.Trichoderma viride TV41 gained the highest sporulation yield of 2.49 × 109cfu/g with the inoculation amount of 10%.Under the condition of triangular flask-culturing,the sporulation yield of Trichoderma viride TV41 was more than the sporulation yield of the shallow pan.Furthermore,regarding the effects of the three different media thickness on the sporulation of Trichoderma viride TV41 in solid fermentation under the condition of shallow pans,the results demonstrated that the sporulation was the highest based on the culture medium thickness of 0.5cm.In the process of generating Trichoderma viride TV41,the thickness of solid culture medium had a great effect on the sporulation of Trichoderma.The thickness of the Trichoderma viride TV41 in the addition of amino acid waste should be controlled in the thickness range of 0.5 cm to 1.0 cm.The promotion effect of Trichoderma viride TV41 on the growth of tomato seedlings under salt stress and the effect on the number of rhizosphere microorganisms were investigated under potted conditions.The results showed that the plant height,root length,stem width,the fresh weight of the ground and underground sections were significantly enhanced in comparison with the untreated groups.However,Trichoderma viride TV41 was ineffectively utilized to the salt disaster in the soil concentration of less than 0.4%.When the soil salt concentration was higher than 0.4%,the salt disaster of the treated groups of Trichoderma viride TV41 was significantly alleviated.Trichoderma viride TV41 can effectively improve the plant growth condition in the soil of a certain salt concentration,and especially under the high concentration of salt stress,the plant growth was more significantly promoted.
