In this study,the waste residue of Ginkgo biloba leaves extracted from economic forest trees with Chinese characteristics was used as raw material,and the high-speed homogenization-ultrasound-assisted extraction method was used to study the green and efficient extraction technology of bilobetin,ginkgetin,isoginkgetin,sciadopitysin in G.biloba leaves residue.A rapid,sensitive,simple and accurate method for qualitative and quantitative determination of four main biflavones in G.biloba leaf residue was established.The effects of various parameters of high-speed homogenization-ultrasound assisted extraction process on the extraction rate were investigated.The optimum technological conditions were obtained.Macroporous adsorption resin was used to enrich the four main biflavones in G.biloba leaves residue.This study realizes the comprehensive utilization of G.biloba leaves residue,provides an important theoretical basis for the study of G.biloba flavonoids,and has important application potential.The results are as follows:1.A method for simultaneous determination of bilobetin,ginkgetin,isoginkgetin,sciadopitysin in G.biloba leaves residue by high performance liquid chromatography was established:Chromatographic column:Phenomenex-Gemini C18 column(250 mm×4.6 mm;5 ?m),velocity of flow 1.0mL/min,column temperature 35?,sample size 20 ?L,detection wavelength 330 nm.The mobile phase was acetonitrile(A)and 0.4%phosphoric acid(B).The gradient elution was performed as follows:0-20 min;55-75%(B).2.The extraction parameters of bilobetin,ginkgetin,isoginkgetin,sciadopitysin from ginkgo G.biloba leaves residue by high-speed homogenization-ultrasound-assisted extraction were optimized,and the optimum parameters of high-speed homogenization-ultrasound-assisted extraction were determined:ethanol concentration:70%,solid-liquid ratio:1:40,homogenization time:2 min,homogenization speed:12×103 r/min,ultrasonic time:20 minUnder these conditions,the extraction rates of bilobetin,ginkgetin,isoglnkgetin,sciadopitysin were 0.10 ng/g,0.14 mg/g,0.11 mg/g and 0.15 mg/g,respectively.The experimental results show that the homogeneous-ultrasound assisted extraction method is a simple,efficient and inexpensive extraction technology,which can be widely applied to the utilization of forest non-wood resources,extracting the efective bioactive components and improving the added value of forest non-wood resources3.Macroporous adsor:ption resin was used to enrich and separate bilobetin,ginkgetin,isoginkgetin,sciadopitysin from G.biloba leaves residue sample solution.The recovery rate of bilobetin,ginkgetin,isoginkgetin,sciadopitysin was obtained.And the recovery rate were 67.85%?71.68%?74.16%?64.51%,respectively.The contents of four biflavones in the extract of G.biloba leaves residue by AB-8 macroporous resin were 1.05%?2.01%?1.62%?1.37%,respectivelyIn summary,the high-speed homogenization-ultrasound-assisted extraction of four kinds of biflavones from G.biloba leaves residue is simple,inexpensive and environmentally friendly.It is suitable for processing and value-added products of G.biloba leaves and other non-wood resources by-products.It helps to improve the quality and efficiency of forest non-wood resources industry and provides technical support for structural reform of forestry supply.
