Obesity is prone to various chronic diseases and has gradually become a health problem that harms human life.Many studies have found that medium-chain fatty acids can effectively improve the lipid metabolism disorder of nutritional obesity,reducing serum's TC and TG levels.The liver is the best place for synthesizing fat,also an important organ for the degradation of cholesterol,which maintains the body's metabolic balance of triglycerides and cholesterol.Bile acids is a vital product of cholesterol metabolism,produced by the degradation of cholesterol and secreted into the small intestine,most of them are reabsorbed back to the liver,and a small part is excreted.MCFA can promote fecal cholesterol and bile acid excretion,which lowers serum cholesterol levels and improves cholesterol metabolism.Cinnamomum camphora seed kernel oil is a natural source of MCFA,which has important research significance.A nutritional obese mouse model closest to human obesity was firstly constructed,then the obese mice were fed with the high-fat diet whose fat was replaced by 100%Cinnamomum camphora seed kernel oil and 50%Cinnamomum camphora seed kernel oil,with obese mice fed high fat diet as model control,and healthy mice fed basal feed as blank control.To explore the effect of Cinnamomum camphora seed kernel oil on ameliorating triglyceride metabolism and lowering Serum Cholesterol in nutritional obese mice,we measured the body weight?body length?body fat and blood lipids,the main organ index and slice,and the excretion of fecal cholesterol and bile acid.The mechanisms of Cinnamomum camphora seed kernel oil on ameliorating triglyceride metabolism and lowering Serum Cholesterol in obese mice was investigated by Real-time PCR and Western blotting,targeting liver and small intestine.The results showed that this kind of oil can effectively reduce the body weight and body fat,lower TG,TC,LDL-C,LDL-C/HDL-Cl,AST and ALT levels in serum in the obese mice,reduce the release of ET from liver to the blood,Improve the index of heart,spleen,lung and kidney,lower liver index,reduce liver and lung damage and protect the health of major organs in obese mice,improving lipid metabolism.Cinnamomum camphora seed kernel oil can significantly promote the excretion of fecal bile acid,including CA,DCA,LCA,CDCA,UDCA,GDCA and TLCA,reducing the accumulation of cholesterol and bile acid,lowering blood cholesterol,thereby improving lipid metabolism.Cinnamomum camphora seed kernel oil can reduce the gene expression of ACC1 and FAS in liver,reducing the onset of fatty acid synthesis.Cinnamomum camphora seed kernel oil can up-regulate the gene expression of LPL in liver,promoting the body to clear lipids and lowering blood lipid levels;Cinnamomum camphora seed kernel oil can up-regulate the expression of intestinal FIAF gene and then inhibit the expression of intestinal LPL gene,Reduce lipid deposition and improve liver triglyceride metabolism.Cinnamomum camphora seed kernel oil can up-regulate the gene and protein expression of liver ABCG8,and promote its translocation of excess cholesterol to bile excretion,reducing the accumulation of cholesterol;Cinnamomum camphora seed kernel oil can downgrade the gene expression level of LXR,and inhibit the expression of SHP gene at the same time,promoting the expression of CYP7A1 gene and protein,strengthening bile acid synthesis.Cinnamomum camphora seed kernel oil can improve the intestinal mucosal barrier integrity by reducing the expression of intestinal TGR5 mRNA to protect the intestinal mucosal barrier to reduce endotoxin in the blood and promote intestinal peristalsis to increase bile acid excretion;At the same time,it has a tendency to down-regulate the expression of IBABP mRNA,reducing intestinal bile acid reabsorption,and thus lowering serum cholesterol.
