Yak bone contains rich collagen,in the process of manufacture of yak meat products,some yak bone is used as food and craft decorations,while the rest is discarded,resulting in waste of resource.In this paper,yak bone collagen is extracted from yak bone as raw material,antioxidant peptide of yak bone collagen is prepared with the radicals scavenging rates of DPPH and OH as indexes,ACE inhibitory peptide is prepared with the inhibitory rate of ACE as indexes,and the prepared yak bone collagen peptide is purified.The specific work content is as follows:1.On the basis of single-factor experiments,taking the extraction rate of yak bone collagen as the evaluation index,and according to the design principle of Box-behnken experiment,the ultra-high pressure coupling acid enzymic method is used to extract yak bone collagen,and the optimum technological conditions are obtained: Degrease with petroleum ether for 2 h,decalcify with 0.5% EDTA solution for 36 h,have ultra-high pressure treatment at 350 MPa for 10 min.The amount of enzyme is 0.4 g/mL,the ratio of solid to liquid is 1:290 g/mL,and the extraction time is 10.5 h.The actual extraction rate is 58.95%.2.Taking the scavenging rate of DPPH· and the scavenging rate of OH· as evaluation indexes respectively,the orthogonal experiment is according to the design principle of Box-behnken experiment of yak collagen antioxidant peptide,and the optimal process conditions are obtained: When the optimal substrate concentration for the scavenging rate of DPPH· is 25 g/L,reaction time is 1.8 h,pH value is 8.0,enzyme dosage is 33 ?L,then we have the optimal radical scavenging rate of DPPH·,which is 32.51%.The best substrate concentration for the scavenging rate of OH· is25 g/L,the reaction time is 1.0 h,the pH value is 8.4,the enzyme dosage is 45 ?L,then we can have the best free radical scavenging rate,which is74.51%.The yak bone collagen is purified by preparative high performance liquid chromatography,and three components are obtained,with the best OH· removal rate of 40.76%.3.Taking the inhibition rate of ACE enzyme activity as the evaluation index,the preparation process of yak collagen ACE inhibitory peptide is and according to thedesign principle of Box-behnken experiment,and the best technological conditions are obtained: Substrate concentration is 10 g/L,reaction time is 1.0 h,pH value is 8.3,temperature is 42?,and the obtained ACE enzyme activity inhibition rate is the best,83.52%.The yak bone collagen ACE inhibitory peptide is purified by preparative high performance liquid chromatography.The ACE inhibitory rate of the simulated digested product is determined.The best optimal component is 75.15%.
