Glucomannans(GMs)from diverse natural plants have great potentiality in enhancing host immune system,protecting intestinal mucosa and regulating gut microbiota.In this study,three kinds of GMs were achieved from Amorphophallus rivieri,Aloe vera,and Dendrobium officinale named KGM,AGP,and DOP,respectively.Dextran sodium sulfate(DSS)was employed to construct the colitis mice model,and three kinds of GMs were administrate to model mice.Effects on gut mucosa injury,intestinal immunity and gut microbiota composition were tested to investigate and compare the anti-inflammatory effects of the three GMs.In addition,effects of GMs on the proliferation of IEC-6 and the viability of inflammatory cells were also tested to evaluate their anti-inflammatory properties in vitro.The main research contents and conclusions were as follows:1.Protective effect of the GMs on gut mucosal injury in DSS-induced colitis mice model were investigated.The model of mice was established by DSS.The three fractions were administrated with or without DSS containing drinking water.The purpose was to investigate the effect of GMs on intestinal mucosal injury in mice.Results showed that administration of the GMs improved the health state of colitis mice,including the recovery of body weight,increase of crypt depth,bowel wall thickness.GMs also alleviated the injury of gut mucosa,promoted the growth of intestinal epithelial cell in colitis mice.In addition,our result showed that the GMs significantly increased proliferation of goblet cell and secretion of mucin proteins,restored the integrity of gut mucosa and function of barrier defense.The above results suggested that GMs can attenuate the intestinal epithelial injury.2.Regulating effects of the GMs on intestinal immunity in colitis mice were studied.The DSS-induced model mice are usually accompanied with intestinal mucosa immunity disorder.The three fractions were administrated with or without DSS containing drinking water.Expressions of related Toll-like receptors were quantified.The results showed that administration of GMs increased immune organ index and immune cells number,and balance the levels of pro-and anti-inflammatory cytokines.The integrity of intestinal mucosa was maintained by the increasing number of goblet cells and mucin proteins production.In addition,the results showed that the GMs significantly increased amount of related immune cells and regulated the expressions of TLR-2,TLR-4,TLR-6,and TLR-9.In conclusion,the GMs could regulate the intestinal mucosal immunity in colitis mice and the possible pathway was dependent on TLRs expression.3.Improvement effect of the GMs on gut microbiota and short chain fatty acids in colitis mice were analyzed.The DSS-induced model mice are usually accompanied with gut microbiota disorder.The three GMs were administrated to colitis mice.Mice feces were collected to investigate the concentration of short chain fatty acids(SFCAs).The different of mice gut microbiota composition caused by three GMs were investigated by gene sequencing method.The results showed that the concentration of SCFAs had significantly increased by the GMs in colitis mice.The abundance of Clostridiales:f_:g_,Ruminococcus,Oscillospira and Flexispira in colitis mice was up-regulated by the GMs,while the abundance of Bacteroides,S24-7,Sutterella and Enterobacteriaceae: g_was down-regulated.These results indicated that the GMs could improve the metabolic environment of intestinal and regulate the imbalance of gut microbiota in colitis mice.4.Protective effect of GMs on cell proliferation and the cell viability of injured IEC-6 cells were explored.DSS was applied to IEC-6 cells to establish a injured intestinal cell model.The three GMs were co-cultured with IEC-6 cells to test their toxicity to cell.The three GMs were co-cultured with injured cell,and the survival rate of the cells was measured.In conclusion,a certain concentration range of GMs were not toxic to cells.The injured cells model of IEC-6 was established by DSS.Additionally,the three GMs improved the cell viability of injured cells.
