Labisia pumila is a traditional herb in Malaysia,The botanical name of Kacip fatimah is Labisia Pumila,a red herb used by Malaysian women for generations.It is a genus of small woody and leafy plants of the Myrsinaceae family,Cubilose,Tongkat Ali,Pewter and Labisia pumila are the four national treasures of Malaysia.At present,the research on Labisia pumila is mainly focused on its water extract,alcohol extract and other crude extract,as for the specific active ingredients and the separation purification conditions of Labisia pumila,no one has reported yet.Flavonoids have strong antioxidant activity,and their antioxidant activity was related to the composition and proportion.Therefore,this paper studied the extraction conditions,components and antioxidant activities of flavonoids from stems and leaves of Labisia pumila,providing a theoretical for its development in the cosmetics industry.At the same time,the absorption characteristics of flavonoids were studied by simulated digestion and intestinal absorption experiments in vitro to provide theoretical for the development and utilization of flavonoids in the health food industry.The main results are as fllows:?1?Response surface optimization?RSM?experiment was used to study the effect of solid-liquid,ethanol concentration,extraction temperature and extraction time on the extration ratio total flavonoids.The best conditions were as follows:solid-siquid,ethanol concentration,extraction temperature and extraction time were selected at 1:60?g/mL?,35%,85?and90min,respectively.Under this condition,the exteaction of flavonoids reached 39.115mg/g DW.The AB-8 macroporous resin was used to separate and purify the crude solution of Labisia pumila.After purify,The leaf flavonoid content?381.20 g rutin/mg DW?increased by nearly 4.5 times;Stem flavonoid content?667.22 g rutin/mg DW?increased by nearly 8.1times.The structures of the purified Flavonoids in the stems and leaves of Labisia pumila were identified by LC-MS,and in S40,11 major flavonoids were identified:catechin>Epicatechin>chrysin>puerarin>rutin>apigenin>epigallocatechin>quercetin>genistein>luteolin>myricetin;in L40,10 major flavonoids were identified:rutin>catechin>eoigallocatechin>keampferol>epicatechin>quercetin>myricetin>genistein>apigenin>puerarin.?2?The antioxidant activities of flavonoids in stem and leaf of Labisia pumila were evaluated by chemical antioxidant method,human hepatocellular carcinoma cell model and human erythrocyte oxidative hemolysis model.The results showed that the antioxidant capacity of flavonoids in stem and leaf of Labisia pumila had significant effects in different antioxidant systems:The flavone leaf extracts performed better than the stem extracts in chemical antioxidative activities,the IC50 value of DPPH and ABTS were 0.99 and 2.26 g Rutin/mL,respectively.The CAA values of S40 and L40 in inhibiting peroxide free radicals were 0.28 and 3.03?mol of Qu/100g powder,respectively,It shows their antioxidative activities were S40>L40.The result of 2,2'-azobis[2-methylpropionamidine]dihydrochloride?AAPH?-induced hemolysis model indicate the protective effects of flavone extracts on erythrocytes against AAPH-induced oxidative stress.In general,the flavonoids from Labisia pumila have strong antioxidant activity.?3?The antioxidant activity of flavonoids in the process of gastric and intestinal digestion was determined by measuring the antioxidant activity of flavonoids after simulated gastrointestinal digestion in vitro.The results showed that the scavenging ability of ABTS free radicals and the reducing ability of Fe3+were not significantly affected while digestion by gastric and intestinal,but the scavenging ability of DPPH free radicals was significantly enhanced after intestinal digestion;Erythrocyte hemolysis test results show that the gastrointestinal digestive system did not significantly alter the antioxidant activity of stem?S40?and leaf?L40?flavonoid,The results of ROS,MDA,GSH,GSH-Px and SOD all showed no significant changes in the process of gastric digestion,but significant changes in the process of intestinal digestion.However,the experimental results at the 6th hour of intestinal digestion were almost consistent with the experimental results at the 0th hour of gastric digestion.?4?Absorptivity of flavone extracts was characterized using a Caco-2 monolayer model,The results showed that the flavonoids of leaves and stems from kacip fatimah had good absorbtion characteristics,while the flavonoids in leaves were easily absorbed in stems.
