The unique optical,electrical,biological and nanoscale properties of gold nanomaterials make them quite popular in a number of research fields including biomedical science.Gold nanomaterials based novel devices exhibit excellent performance in biomolecule detection.Gold nanomaterials related preparation methods are normally divided into chemical and physical approaches.In this article,we have prepared the plasmonic gold chip with special optical property through solution phase chemical reduction and explored its application in biomolecule detection.The plasmonic gold chip exhibits remarkable fluorescence enhancement of about two orders of magnitude in near-infrared region compared to glass chip.When integrated with high-throughput microarray technique,the plasmonic gold chip has been used for detection of three typical lung cancer markers,namely CEA,Cyfra21-1 and NSE.In comparison with the Luminex platform,lower detection limit and quantitation limit have been achieved by using our chip due to the better stability at low concentration.Through properly adjusting the threshold,we've improved the sensitivity under high specificity.ROC curves also suggest the superiority of the plasmonic gold chip in detection of the three markers.Combination of these markers gives better performance than any one marker alone with the sensitivity of 52.08% and the specificity of 95.83%.The plasmonic gold chip based combined detection of tumor markers will provide considerable information for lung cancer diagnosis and have broad applications.
