For the prevention and treatment of some diseases,it is necessary to delivery subunit vaccines and DNA vaccines in the form of nanoparticles to the cortex.Microneedle is kind of array patch with needles less than 1 millimeter.As an effective cortex-targeting delivery system,whether microneedle load nanoparticle therapeuticals and deliver to cortex is the main aim of this project.As emerging therapeutical drugs,microneedle is not only a painlessness and easy-adapting transdermal system,but also can promote the absorption and targeting of water soluble drugs and their nanoparticles,and avoid gastrointestinal digestion and first-pass effect.Hopefully,the combination of mironeedle and nanopaticle drugs could create a new applicaion field.In this project,we expected to use microneedle to carry nanoparticle drug model through soaking and observe the distribution of model drug on needle body and inwaird diffusion.We modified the prescription of microneedle on the basis of the phase-transition hydrogel microneedles invented by our lab through application of hyaluronic acid and CMC-Na to maximize the swelling of microneedle.Meanwhile,quantum dots are used as drug model to explore feasibility of loading nanoparticle drugs on transdermal microneedle patches.Then,we make sodium alginate degradate through acid and oxidation to get sodium alginate with small molecular weight and expect to form nanoparticle between sodium alginate and PEI through electrostatic interactions.We expected to synthesize blocks by benzaldehyde and serine to wrap the ploycationic nanoparticle to decrease the toxicity of polycationic particles.In order to test the applicability of this soaking method,we dipped phase-transition microneedle into fluorescent liposome and analyzed the result qualitatively.
