Traditional wound dressings are gradually unable to meet the increasing need of healing wounds caused by accidents or illness.Uncontrollable bleeding has been the main factor causing death.Natural macromolecular dextran is of quality that can promote wound healing and dextran-aldehyde formed by oxidizing dextran is biocompatible,anti-bacterial and able to be used to produce compound wound dressing or hemostatic agent with other polymers.Therefore,developing hydrogel sponge wound dressing and hemostatic is of great value of application.PVA/DA hydrogel sponges were prepared by blending different ratios of Poly?vinyl alcohol?/Dextran-aldehyde solution,followed by crosslinking via freeze-thaw method and freeze-drying.PVA/DA hydrogel sponge showed can absorb fluid of over 6 time its weight and structures remain stable.The hydrogel also showed suitable water vapor transmission rate of 2100 g·m-2·day-1 to keep a moist environment and good tensile strength and flexibility.SEM proved 3-dimensionnal porous internal structure,with pore sizes ranging from 510?m.Good biocompatibility was proved by very low hemolysis and no cytotoxicity.Full-thickness wound healing tests prove the hydrogel sponge can promote faster regeneration of skin and wound healing.Thus,the PVA/DA hydrogel is suitable for application as a wound dressing.Gel/DA hydrogel sponge was prepared by blending gelatin and dextran-aldehyde solution and freeze-drying.The Gel/DA hydrogel sponge can quickly absorb 4 time its weight water within 5 seconds before reaching plateau of 8.5 time and remain structurally stable after72 h.SEM images showed a highly interconnected porous and lamellar structure favorable for forming clot,with pore sizes ranging from 100200?m.In vitro pro-coagulant test proved that Gel/DA sponge can reduce blood coagulation time to 170 s by accumulating red blood cells on the surface,further proved by SEM images of adhered erythrocytes.Rat hemorrhage test results showed Gel/DA sponge can reduce hemostatic time to 65 s and blood loss to 0.66g,while no liver damage or inflammatory reaction was observed.In addition,Gel/DA sponge could promote full-thickness wound healing.Thus,Gel/DA hydrogel sponge showed potential for use in hemostatic management and wound healing.
