In the first part,molecularly imprinted polymers(MIPs)are materials with specific recognition properties that synthesized by molecular imprinting technology(MIT),which shows great potential applications in drug delivery systems.However,most MIPs have the disadvantages of low drug loading capacity,easy collapse or low bioavailability,which limit their applications in drug delivery system as carriers.In order to solve these problems,on one hand we added liquid crystal monomer which could decrase reduse crosslinking degree to improve the rigid structure of polymer molecules,and on the other hand,added polyhedral oligomeric silsesquioxanes(POSS)with cage structure to increase its hydrophobicity,and constructed a liquid crystalline and polyhedral oligomeric silsesquioxanes-based molecularly imprinted drug delivery system with superior floating nature.Its'density was lower than the gastric juice(??1g/cm~3),so it can stay floating in the stomach without affecting the rate of gastric emptying for a long time,thus the residence time in the stomach was prolonged and could be used for the studies of sustained release of capecitabine.This preparation method was simple and rapid.Through the preparation condition optimization,characterization experiments and in vitro and in vivo release studies,the great advantages of this imprinted material were proved.Molecularly imprinted polymers were prepared by using capecitabine as the template,methacrylic acid as the functional monomer,and ethylene glycol dimethacrylate as the crosslinker,toluene and acetonitrile as the porogens and synthesised by bulk polymerization.Compared with the control groups,this material had good floatation performance in vitro and in vivo,and showed good controlled release effect of capecitabine and higher bioavailability in the pharmacokinetic study of rats in vivo.These results indicated that the molecularly imprinted gastric retention drug delivery system based on liquid crystal and POSS had a great development prospect in the new formulation of capecitabine for sustained and controlled release.The second part focused on the research importance of current proteomics.It was more and more prominent to develop new composite materials as adsorbents for solid phase extraction to separate and enrich proteins.The key of this technology was to design functional solid phase extraction fillers.Due to its unique optical,electrical and magnetic properties,gold nanorods have considerable application prospects in nanomaterials.Among them,the adjustable aspect ratio was particularly remarkable,which greatly expands its application fields.Related studies had reported that gold nanorods had good binding capacity for bovine serum albumin(BSA)when the aspect ratio is 3-8.However,the exposed gold nanorods tended to aggregate and change their morphology.Improper modification can also weak the activity of protein.Therefore,we designed a green solvent room temperature ionic liquid to synthesize the polymer material,and introduced the gold nanorods into the nanocomposite to obtain uniformity and dispersed extraction fillers,it was expected to optimize the preparation conditions and extraction conditions to achieve rapid separation and enrichment of albumin in complex samples,providing a new idea for the separation and purification of proteins.Using green solvent ionic liquid as the porogen,glycidyl methacrylate(GMA)as the functional monomer and ethylene glycol dimethacrylate(EDMA)as the crosslinker,the polymeric material was synthesized under the action of initiator2,2'-Azobis(2-isobutyronitrile).The gold nanorods can be grafted successfully by the amino reaction of the material.The characterization experiments showed that the material had been successfully grafted with gold nanorods and has a uniform pore structure.The extraction ability of bovine serum albumin was up to 99.80%and the reproducibility was good.It can separate bovine serum albumin and cytochrome C successfully and enrich albumin in normal human serum.The results showed that gold nanorods grafted polymer which prepared by ionic liquids could significantly improve the ability of protein adsorption and separation and enrichment of bovine serum albumin(BSA).With the further study of the composite material,it could play a more and more important role in the field of protein analysis and separation.
