As the biggest killer for female cancer,breast cancer with the highest incidence and mortality has been the greast threat for female health all over the world.In recent years,the diagnosis and combination therapy of breast cancer have been the focus of nanomedicine research.With the rapid development of biomaterials,nanotechnologies,and clinical cancer treatment,revolutionary advances have been made for the breast cancer precise theranostics based on nanomaterials.The aim to develop the multifunctional and multimodal agents with both diagnostic and therapeutic properties based on nanomaterials has proved to be one of the efficient strategies for the diagnosis,imaging and treatment of breast cancer.In this study,based on the iron oxide nanoparticles?IONPs?with simple preparation process,low synthesis cost and excellent biological performances,we have developed dual-drug-loaded magnetic hydrogel?DDMH?for alternative magnetic field responsive synergistic chemotherapy,manganese-doped magnetic layered double hydroxide nanopletes for both T1-and T2-weighted enhanced magnetic resonance imaging?MRI?,and drug-loaded magnetic nanocubes as multifunctional platform for theranostics of breast cancer.Meanwhile,after the evaluation of physicochemical properties and biocompatibility of as-synthesized agents,the feasibility and effectiveness of the three theranostic agents were further systemly evaluated using both in vitro cancer cells and in vivo tumor-bearing mice.The results demonstrated that Doxorubcin and Doxtaxel in DDMH exhibited excellent synergistic inhibition on the growth of both triple negative breast cancer cells and tumor of tumor-bearing mouse modal.Furthermore,the asynchronous release behavior and alternative magnetic field responsive realese property of DDMH could further enhance the inhibition effect.Both in vitro and in vivo MRI measurement showed that MgMnAl-LDH@IO NPs have excellent both T1-and T2-weighted enhanced MRI contrast effects,which further provide more accurate pathological information about tumor tissues.Besides providing the T2-weighted MR imaging information,the IOMS-PEG?DOX?-2DG NCs could combine the photothermal therapy and chemotherapy to realise the goal of both in vitro and in vivo targeted breast cancer theranostics,with the guide of active targeting agent 2-DG.
