There is a high incidence of diabetes in the world.Severe diabetes patients are requested to receive injection of insulin to control the blood glucose concentration in the last stage of diabetes.Even so,injection of insulin is still not an ideal treatment for diabetes.Patients' blood glucose concentration changes dramatically due to impaired insulin release and control system in vivo.Acute blood glucose fluctuations usually bring an additional damage to human health.Occasionally,a lethal hypoglycemic shock occurs to those diabetic patients with insulin treatment.Therefore,it's the key point of diabetes treatment for stabilizing the blood glucose and avoiding a hypoglycemic event.But the traditional treatment,“blood glucose monitoring and insulin injecting” can ‘t monitor glucose in a real time and inject a moderate insulin to stabilized glucose concentration.This project tried to design a brand-new insulin delivery system,not the traditional treatment “insulin injecting after glucose monitor”,but using glucose responsive material to achieve the goal that stabilizing blood glucose.In this project,article researching had been done,firstly.And a new nano-scale polymer material named PAM-PAspPBA-PEG was designed.insulin can be loaded in the core of polymer by material self-assembly.And the polymer is glucose-responsibility which means it can release insulin through mimic physiological insulin release system when the glucose changing.It's a closed-loop system which can manage insulin releasing dosage and keep blood glucose staying in a normal level.After material synthesized,we completed series of identification experiments,such as TEM,DLS,and HNMR to measure the physicochemical properties of the material,including size,molecular composition and configuration.In the latter part of project,several experiments were used to verify the glucose-responsibility of insulin loading polymer and whether it can stay blood glucose in normal level.Besides that,we also built up a diabetes animal model to test the glucose-responsibility,effectiveness and safety in vivo through the insulin release ratio of polymer in different glucose concentration.After all those experiments,results show that the polymer,PAM-PAspPBA-PEG,is glucoseresponsive material in Nano-scale.It also was effective and safety in diabetic animal compared with regular insulin injection.Most of these results are basic what we expected.In summary,we synthesized a self-assembly Nano-scale material,PAM-PAspPBA-PEG,which can release insulin when glucose level rising.This polymer delivers insulin safety and effective through detecting body glucose changing without additional complicated blood glucose monitoring system.So,it has a good novelty and could be a promising application in diabetic treatment.
