Antarctic krill(Euphausia superba)is a single species of plankton that lives in the Central South Ocean in a cluster.It has a huge biomass.Antarctic krill has become the most strategic resource in China.Important development object.N-3 LCPUFA such as EPA/DHA in Antarctic krill oil is more in the form of phospholipids,and has better bioavailability and physiological activity.Therefore,to explore the nutritional mechanism of Antarctic krill oil on the improvement of lipid metabolism in the body,and the impact on the bioavailability of other fat-soluble components astaxanthin,to provide scientific research for its development and application in the field of functional foods or special medical foods.Basis and theoretical support.Experiment 1:Effects of feeding Antarctic krill oil on the lipid metabolism in ratsObjective: To investigate the effects of feeding Antarctic krill oil on lipid composition,especially fatty acid composition,in target organ tissues(serum,liver)of rats.Methods: Male Wistar rats were randomly divided into 4 groups: blank control group,soybean oil group,triglyceride type fish oil group and Antarctic krill oil group.The intragastric dose was 2.625g/kg,continuous gavage for 30 days.Changes in serum triglyceride(TG),cholesterol(TC)levels,serum and liver fatty acid composition in rats.Results: Compared with the control group,the serum TG content of the Antarctic krill oil group and the fish oil group decreased by 19.0% and 9.52%,respectively,and the TC content decreased by 8.9% and 4.14%,respectively.The serumEPA content increased by 309%(P<0.05),340%(P<0.05),DHA content increased by 80.20%(P<0.05)and 42.32%(P<0.05),respectively,AA content decreased by 37.67%(P<0.05),42.19 %(P<0.05);The EPA content in the liver increased by 591.43%(P<0.05)and 548.57%(P<0.05),and DHA content increased by 48.12%(P<0.05)and 42.26%(P<0.05),AA content decreased by 45.77%(P<0.05)and 33.63%(P<0.05).Conclusion:Antarctic krill oil can improve lipid metabolism and increase serum and liver EPA and DHA levels,and the effect is better than triglyceride type fish oil.Experiment 2:Effect of feeding Antarctic krill oil on digestion,absorption and utilization of dietary astaxanthinObjective: To establish an assay for astaxanthin in different tissues and organs of animals,and to explore whether Antarctic krill oil has an effect on the digestion and absorption of astaxanthin,and provide for the development and application of Antarctic krill oil in functional foods or special medical foods.Scientific basis and theoretical support.Methods: 210 Wistar male rats were randomly divided into 5 groups: blank control group,krill oil group(administered dose 0.208g/kg),astaxanthin group(administered dose 1.25mg/kg),shrimp green.Suspension + fish oil group(including astaxanthin 1.25mg/kg,fish oil dosage 0.208g/kg)and Antarctic krill oil + astaxanthin group(including astaxanthin 1.25mg/kg,shrimp)Oil perfusion dose 0.208g/kg),anesthetized with sodi?m 3% pentobarbital(0.3ml/100g)at 0h,0.5h,1h,2h,4h,8h,10 h,12h,16 h,24h after gavage The abdominal aorta was sacrificed after blood was taken,the serum was separated,the small intestine(stomach cardia to the cecum),the contents of the small intestine,the small intestine villi,and the astaxanthin content in the tissues and organs were detected.Results: Methodology validation meets the requirements for astaxanthin analysis in tissue organs.Astaxanthin ester enters the rat body.The concentration of astaxanthin in the serum of each group slowly rises at 0.5h after gavage,reaches a peak at 8h,and the highest concentration in serum is 4.02 times that of fish oil + astaxanthin group.8.37 times the green group.The area under the astaxanthin curve(AUC)of the Antarctic krill oil + astaxanthin group,astaxanthin group and fish oil + astaxanthin group was 108.39±2.34?g?h/L,14.35±0.21?g?h/L,23.39±0.46?g?h/L,reflecting that the bioavailability of Antarctic krill oil and astaxanthin in rats is higher than that of the astaxanthin group,which is superior to fish oil and astaxanthin.Ester compounding.The astaxanthin Antarctic krill oil + astaxanthin group in the intestinal contents of rats reached the peak at 4 h,delayed by 2 h than the astaxanthin group,and 7.40 times that of the astaxanthin group.Astaxanthin Antarctic krill oil + astaxanthin group,fish oil + astaxanthin group,astaxanthin group reached the peak at 4h,and the highest concentration was 3.09 times of fish oil + astaxanthin group.The green pigment group was 6.88 times.After 12 h,the content of free astaxanthin changed slowly.By 24 h,the free astaxanthin content was basically empty in the small intestine.Conclusion: The serum absorption rate and organ tissue accumulation rate of the Antarctic krill oil and astaxanthin ester combination group are excellent.Antarctic krill oil can promote the bioavailability of astaxanthin,which provides scientific basis and theoretical support for the development and application of functional foods or special medical foods.
