Fluorescence probe technology refers to the use of certain features of fluorescence,and the fluorescence(excitation wavelength and emission wavelength,intensity,lifetime,polarization etc.)with the nature of the environment,a class of molecules such as polarity,refractive index,viscosity change and change sensitive compounds,research on some special environment materials or the target material structure,content and some other physical or chemical properties of the molecular level qualitative or quantitative technology,it also has the characteristics of high sensitivity and wide dynamic stability of the corresponding time.With the continuous development of physical and chemical synthesis technology,through the control of fluorescent molecules in the process of transferring information,subject to different environmental stimuli to achieve open and close the fluorescence conversion,thus more accurate in vivo protein molecules and the structure of other ions were identified and marked,and makes quantitative detection of them more accurate.As a probe in the field of life science,environmental science,materials science,information science and other fields has been widely used in recent years.This paper is mainly based on the,thiols on beta unsaturated nucleophilic addition,through the design and synthesis of a 3-amino phenol as the main raw material,the synthesis of 7-amino-4-methyl coumarin as a fluorescent group,using maleimide or its derivatives as acceptor,specific fluorescent probe for detection of biological thiols.The optimization and yield of each step reaction in the synthesis process were studied and analyzed,and the synthesis conditions were optimized.And the fluorescence probe out a series of studies for the detection process,analysis in different environments,selectivity and stability of thiol content detection simulation of intracellular environment influence under the condition of common impurity.Research status and prospects of development combined with the importance of biological thiols and detection methods,and expand their synthesis of fluorescent probe for the experimental work,the optimization of synthesis of fluorescent probe and the detection results and analysis of biological thiols.
