Dihydromyricetin?DMY?is a special flavonoid with many physiological functions such as anti-microbial,anti-inflammatory,analgesia,anti-oxidant and anti-tumor.The special molecular structure of DMY makes it possible to form stable complexes with metal ions.In the present work,DMY-Zn???,DMY-Mn???and DMY-Ni???were synthesized by heating reflux method,and the structure was characterized by the technology method.Prediction of binding sites by quantum chemical calculation.The stability,the ability to scavenge hydroxyl radicals and in vitro cytotoxicity of the complexs were studied.The main results and conclusions of this study were described as follows:1.DMY-Zn???,DMY-Mn???,DMY-Ni???complex were synthesized at the condition of70°C,pH=7.5-8,reaction time 8 hours.The most possible structure of complexes are[C15H10O8Zn]·2H2O,[C15H10O8Mn·2H2O]·2H2O,[C15H10O8Ni]·4H2O by the methods of UV-Visible spectrum,IR,TG-DTA and elemental analysis.2.Using quantum chemical method to predict the reactive sites and coordination sites of DMY.The most possible antioxidant active site is C4,,phenolic hydroxyl in DMY molecule is the highest;When DMY complexes with metal ions,the most likely coordination sites are 5-hydroxyl and 4-position carbonyl.3.The stability of the complexes was studied.The results show that the DMY-Zn???,DMY-Mn???and DMY-Ni???complexes have good stability in an alkaline environment and heating within 100°C for less than 100 min.4.The ability of DMY-Zn???,DMY-Mn???and DMY-Ni???complexes to scavenge hydroxyl radicals was studied.The results showed that the ability of the three complexes to scavenge hydroxyl radicals was stronger than that of DMY.5.The effects of DMY-Zn???,DMY-Mn???and DMY-Ni???complexes on the L-02 and AML12 were studied by MTT assay.The results showed that DMY-Zn???,DMY-Mn???,DMY-Ni???complex and DMY inhibited the growth of L-02 cells and AML12 cells in a dose-and time-dependent manner.At low concentration and short time,DMY-Zn???,DMY-Mn???and DMY-Ni???complexes had no obvious inhibitory effect on the growth of L-02 cells and AML12 cells;At high concentration and long time,the inhibitory effect of complexes on L-02 cells and AML12 cells was stronger than that of DMY.
