In this paper,deer oil was extracted from fresh deer?Cervus nippon?fat for further study.The extraction rate of deer oil was used as an indicator by optimizing the optimal process parameters of supercritical CO2 extraction,constant temperature heating,enzymatic hydrolysis and subcritical extraction.The fatty acid composition of the deer oil extracted under the optimal conditions was detected by gas chromatography-mass spectrometry?GC-MS?,and the antioxidant activity of deer oil was also determined.Finally,the application of deer oil for saponification in the field of daily chemical had been indentified.The main results of this paper are as follows:1.Using supercritical CO2 extraction method,constant temperature heating method,enzymatic hydrolysis method and subcritical extraction method,these four different methods were used to extract deer oil from deer fat,and determined the optimum process parameters for each extraction method.Based on comparing the extraction rates of the four methods,then observed and contrasted the sensory factors of deer oil,such as the state,color and smell.Finally,it was determined that the deer oil obtained by the supercritical CO2 extraction method has high extraction rate and good quality,and it was more suitable for further research and analysis.2.The components of deer oil were detected by gas chromatography-mass spectrometry?GC-MS?and sixteen kinds of fatty acids were determined.The relative contents of saturated fatty acids and unsaturated fatty acids were 69.8%and 30.2%respectively.According to the analysis of the properties of various fatty acids,it is determined that deer oil has the functions of emulsification,water retention and anti-oxidation,and has certain development potential in the field of daily chemical industry.3.For studying the antioxidant activity of obtained deer oil,it was learned that the scavenging rate of deer oil on DPPH free radicals was 14.3%,on hydroxyl radicals was6.8%,and on superoxide anion radicals was 4.7%.According to these data,it is demonstrated that deer oil has certain antioxidant activity.4.The extracted deer oil was saponified according to different ratios to optimize the saponification parameters.The results showed that the optimal saponification parameters as the amount of sodium hydroxide was 1314%of the mass of deer oil,and the amount of water was 2.6 times the mass of sodium hydroxide.In order to optimize production process,it is demonstrated that the cream soap base and the saponified deer oil were mixed in a ratio of 3:1,and the optimized ratio was more economical and effective.When the amount of the hand-made deer oil soap flavor was 1.00‰1.50‰of the mixed liquid mass,the cleaning effect was significantly optimal.
