Paclitaxel is a broad-based anticancer drug that can effectively treat many types of cancer including ovarian cancer,lung cancer,breast cancer and non-small cell lung cancer.Paclitaxel is isolated from the slow-growing yew bark.The supply is very limited and the cost of production is high.The separation of paclitaxel from natural yew trees is environmentally unsustainable.At present,there are problems such as low recovery rate in the separation and purification process of paclitaxel,large waste of solvent in the process of separation and purification,and environmental pollution.In addition,the controlled release characteristics of natural drug delivery vehicles and biodegradability are the focus of research.In response to the above problems,this paper developed a novel process for the separation and purification of paclitaxel,and developed a novel drug delivery carrier of paclitaxel while obtaining high-purity,high-yield paclitaxel,which can trigger drug delivery through pH sensitivity.In order to achieve better cancer treatment effect,the drug delivery sustained-release carrier is also combined with photothermal therapy,and the designed carrier is combined with photothermal therapy at the same time as drug treatment.The main contents of this thesis include:(1)Separation of paclitaxel from natural yew trees and rough extraction.(2)The crude paclitaxel sample obtained in the previous step is refined to make the purity of paclitaxel reach the medicinal standard.(3)Construct a controlled sustained-release carrier,and the constructed sustained-release carrier has a photothermal effect at the same time,which can achieve better cancer treatment effect.
