Biogenic amines(BAs)are low molecular weight organic nitrogenous compounds.BAs of aquatic foods are formed microbial decarboxylation of free amino acids.However,the not fresh or even corrupt aquatic foods have a high content of BAs,especially histamine,human consumption can cause allergic symptoms,serious or even lead to death.Therefore,in this study,a pre-column derivatization with dansylchloride reversed high performance liquid chromatography(RP-HPLC)method was developed for rapid determination of BAs in seafood.The species of BAs-forming bacteria in Cololabis saira were isolated and identified,and their abilities of producing BAs.And the effects of storage temperature and bio-preserving agent on the content of biogenic amines in Cololabis saira were studied to find the effective control method of Cololabis saira BAs in the storage and processing.RP-HPLC method was developed for rapid determination of BAs in seafood.Under the optimized experimental conditions,Tryptamine,2-Phenylethylamine,Putrescine,Cadaverine,Histamine,Tyramine,Spermidine,and Spermine have been effectively separated within 15 min.It was linear in the range of concentrations between 1 and 100 mg/L(R~2?0.999).The limits of detection were 4.0?50.0?g/L,and the average recovery of biogenic amines with different mass concentrations were between 79%?117%.The 8 species of BAs in 5 kinds of aquatic products were analyzed by this method,the results showed that the total content of BAs was between23.02 and 228.4 mg/kg.Cololabis saira was selected as raw materials because it is easy to produce the histamine in storage,and a total of 14 strains of BAs-forming bacteria were isolated and purified from the Cololabis saira by using the BAs-forming bacteria screening medium(the modified Niven's agar medium).The 14 strains of BAs-forming bacteria were identified by 16S rDNA sequence and VETIK2 Compact system as Morganella morganii,Klebsiella oxytoca,Enterobacter asburiaede,Citrobacter freundii,Raoultella planticola,Kluyvera ascorbata,Serratia marcescens,Enterobacter gergoviae,Enterobacter sp.,Proteus sp.,Kluyvera intermedia,Proteus penneri,Providencia rustigianii,Hafnia alvei,respectively.Gram staining showed they were all gram negative bacteria.Their abilities of producing BAs at different temperatures(4?,25?,30?,37?)were measured.And find that 6 strains of BAs-forming bacteria has a strong ability to produce histamine(7431.5?10891.8 mg/L)at 25?,30?,37?.But at 4?under the conditions of culture,all BAs-forming bacteria produced histamine very low(ND?26.95)mg/L,and the production of other BAs content is also very low.By investigated the changes of the content of the BAs in the Cololabis saira at different temperatures(-30?,4?,25?),and explored the efficacy of Gracilaria lemaneiformis oligosaccharide,chitosan and the combination of Gracilaria lemaneiformis oligosaccharide and chitosan on the BAs,K value,and total volatile basic nitrogen of refrigerated Cololabis saira fillets.The results showed that low temperature storage can effectively inhibit the formation of biogenic amines in Cololabis saira.Compared with the control,the different treatment with Gracilaria lemaneiformis oligosaccharide,chitosan,and Gracilaria lemaneiformis oligosaccharide-chitosan prolonged the shelf life of Cololabis saira 1 d,4 d,and 5 d,respectively.And the content of BAs in the treated group was significantly lower than the control.It showed that the Gracilaria lemaneiformis oligosaccharide-chitosan could significantly inhibit the production of BAs and prolong the shelf life of Cololabis saira.
