In this paper,pepper essential oil and clove essential oil were selected as experimental materials,and their chemical compositions were determined by gas chromatography–mass spectrometry.Staphylococcus aureus ATCC 25923,Staphylococcus aureus CMCC 26003,Escherichia coli ATCC 25922 and Escherichia coli O157:H7 NCTC 12900 were used as experimental strains to study the antibacterial effects of the two essential oils.Staphylococcus aureus ATCC and Escherichia coli O157:H7 with good bacteriostatic effect were selected as test bacteria to study the antibacterial mechanism of the two essential oils,and the two essential oils were applied to the inoculated ham.The antibacterial and antiseptic effectof essential oil were determined.The research results were as follows:1.GC–MS was used to analyze the main components of essential oil of pepper and clove.A total of 36 substances were detected from the essential oil of pepper,of which linalool was the main component,and the relative mass fraction was 68.011%.A total of 31 substances were detected in clove oil,of which eugenol was the main component,and the relative mass fraction was 76.737%.2.Analysis of antibacterial activity of pepper oil and clove oil.Through the antibacterial circle experiment,the minimum antibacterial concentration(MIC),and the minimum bactericidal concentration(MBC)comparison,we found that essential oils have different inhibitory effects on different pathogenic bacteria.The inhibitory effect of pepper essential oil on escherichia coli is greater than that of staphylococcus aureus.The inhibitory effect of clove essential oil on staphylococcus aureus and escherichia coli is basically the same.The inhibitory effect of clove essential oil on staphylococcus aureus is stronger than that of pepper essential oil.Bacterial growth curve showed that the treatment of essential oil delayed the time of bacterial growth,and bacterial growth was inhibited.3.Study the antibacterial mechanism of pepper essential oil and clove essential oil.Through the observation of transmission electron microscope,the determination of cell content leakage and the determination of cell membrane integrity,we found that after the treatment of bacteria with essential oils,the structural integrity of bacterial cell membranes was destroyed,and the content of cells was leaked,affecting the normal life activities of bacteria,and eventually inhibits their growth and even kills them.4.Analysis of practical antiseptic and bactericidal application of essential oils.The effects of pepper essential oil and clove essential oil on bacterial growth were studied by using liquid medium and ham sausage as culture matrix.Under liquid medium conditions,1MIC,2 MIC and 3 MIC pepper essential oil and clove essential oil can inhibit or even kill bacteria within 120 min;As a culture matrix,the ham intestine was treated with essential oils.The number of bacterial survival in the ham intestine was significantly reduced,which was significantly lower than that without essential oils,indicated that 10 MIC essential oils can inhibit bacterial growth and reproduction within 72 h,but they can not completely kill bacteria.
