| ObjectiveTo develop a complex consisting of grape seed extract,natural vitamin E and selenium enriched yeast the optimal ratio.Then,animal and human experiments were combined to systematically study its edible safety and antioxidant effect,so as to provide scientific basis for its development and application as a compound antioxidant preparation,and to develop and utilize three natural antioxidants scientifically Antioxidants provide important research basis.Methods1.Antioxidant capacity of different combinations of grape seed extract,natural vitamin E and selenium enriched yeast was explored,and the best ratio was selected to make a complex consisting according to the production process requirements of health food.Functional ingredient content,hygiene index and stability of the complex consisting were measured.2.Toxicological safety of the complex consisting was investigated by acute oral toxicity test in mice,three genotoxicity tests and rat 30-day feeding test.3.The aged NIH mice were continuously administered with different doses of the complex consisting for 30 days to explore the effect on antioxidant capacity of aged mice.4.The people aged 40-65 were selected to take the compound preparation for 3 months continuously to observe its effect on the antioxidant capacity of human body.Results1.Grape seed extract,natural vitamin E and selenium enriched yeast have the strongest antioxidant capacity when combined in a ratio of 20:3:2.Proanthocyanidins,vitamin E,selenium contents and hygienic indicators of the complex consisting prepared by that ratio remained stable during the 3-month accelerated test,and its hygienic indicators are in line with the requirements of national food safety standards.2.The value of LD50 of grape seed extract,vitamin E,selenium complex was greater than 20.0g/kg BW,which belongs to the non-toxic grade;Results ofmicronucleus test of bone marrow cells in mice,mouse sperm deformity test and Ames test demonstrated that the complex has no mutagenic effect;30-day feeding test showed that the complex consisting did not affect body weight,food utilization rate,organ weight,blood routine,liver and kidney function,glucose and lipid metabolism of SD rats,and the gross anatomy and histopathological examination of the main organs of rats were also normal.3.After a 30-day administration of grape seed extract,vitamin E and selenium complex consisting,serum MDA and protein carbonyl content of mice in the 0.10 and 0.30g/kg BW dose groups decreased significantly(P<0.05),while GSH,SOD and GSH-Px were significantly increased(P<0.05).4.After 3 months of continuous administration of grape seed extract,vitamin E and selenium complex consisting at 0.62g/d,the self-comparison of the experimental group before and after,and the comparison with the control group,the serum MDA content was significantly decreased and the activities of SOD and GSH-Px were significantly increased(P<0.05),the decrease rate of MDA content,the increase rate of SOD activity and the increase rate of GSH PX in the test group were also significantly higher than those in the control group(P<0.05).Conclusion1.The antioxidant ability of grape seed extract,natural vitamin E and selenium enriched yeast has obvious advantages when they are compatible at the ratio of 20:3:2.The hygienic indexes and stability of grape seed extract,vitamin E and selenium complex consisting prepared by this ratio are good.2.Grape seed extract,vitamin E,selenium complex consisting has good food safety,and can significantly improve the antioxidant capacity of aged mice.3.Daily supplementation of 0.62g grape seed extract,vitamin E and selenium complex consisting could significantly enhance the antioxidant capacity of people aged 40-65 years.4.Grape seed extract,vitamin E and selenium complex consisting are suitable for development and application as a compound antioxidant. |
