At present,the production method of fumaric acid mainly relies on the petroleum-based chemical method,and the bio-fermentation production of fumaric acid can overcome the shortage of non-renewable raw materials and is more environmentally friendly.By the difference of medium concentration and viscosity,the seeds can be cultured into small spherical mycelium with uniform size.After fermentation for 132 h at 200 rpm and 30?,the highest concentration of fumaric acid can reach 33.8 g/L.By using the immobilization of Rhizopus arrhizus mesh and the immobilization of loofah,the fermentation cycle is shortened by 36.369%and the yield is improved compared to the free globular fermentation.This experiment produces rich horses by using glucose as a substrate.Acid,optimized for optimal substrate concentration,fermentation in a 5 liter fermentor(3 liters)through 30? 2 vvm aeration,400 rmp agitation speed,100 g/L monohydrate glucose,can get a conversion rate of 31.42%.By using the COPASI software for modeling and fitting,After fitting the five sets of data,the average R2 value is greater than 0.9,indicating that the fitting effect is good.the key substances affecting the production of fumaric acid,pyruvate and oxaloacetate,and the six-step key metabolic reaction were found.Through the analysis of nitrogen sources,the effects of six common organie nitrogen sources on fermentation were investigated.The results showed that casein can obtain higher space-time yield of fumaric acid;peptone can obtain higher conversion rate,yeast Dip powder can obtain higher biomass due to the richness of nutrients contained in it.At the same time,the fermentation process was adjusted.Compared with the improved one-step culture and the stepwise culture method,similar yields were obtained,and 23.07%of the carbon source,49.75%of the nitrogen source and nearly 100%of the water were saved.Dosage.The nitrogen source in the one-step culture method was compared in terms of types and addition amounts.Studies have shown that the optimal fermentation concentration of 2 g/L in one-step culture with peptone as a nitrogen source can yield 45.84 g/L fumaric acid production.The yield of fermented fumaric acid was further increased by 35.62%compared with the optimized free state.Fumaric received by the solubility and pH of the large effect of temperature,and therefore their properties can be obtained using the extraction rate of 80.33%of a secondary separation fumaric by cooling crystallization and separation of the activated carbon adsorption,fumaric final The purity can be greater than 95%.
