As is known to all,influenza is a global infectious disease,threatening the life and health of human.As a result,the drug therapy for influenza viruses needs to start within 48 hours of onset,this requires a more rapid,ready and convenient detection method.Due to the advantages of high sensitivity,low detection limit,good selectivity and simplicity of the fluorescent probe,we chose it to detect influenza viruses rapidly.Owing to the neuraminidase(NA)encapsulated on the surface of influenza virus having the ability to hydrolyze sialic acid glycoside bonds,we designed and prepared glycosylated fluorescent probes with sialic acid as the major material.In this study,we synthesized fluorescent probe molecules SA-SA.Because of the fluorescent probe involving two effects which are excited-state intramolecular proton transfer(ESIPT)and aggregation-induced emission(AIE),the probe has the advantages of strong luminescence,large Stokes displacement and good light stability.The main purpose of synthesizing two control molecules Gal-Gal and Glu-Glu is to prove the effectiveness and sensitivity of SA-SA.In addition,the synthesis steps of the probe are brief and the major materials are cheap and readily available.In terms of the detection of influenza virus,we incubate influenza viruses with the fluorescent probes,then,we use fluorescence spectrophotometer and microplate reader to detect the fluorescence intensity.This does not involve the tedious biological experiments,the detection steps become very convenient and simple.To improve the accuracy of the experiment,we replace influenza viruses with high purity NA.Meanwhile,we replaced NA with drug-resistant NA to design method that allows our fluorescent probes can be used for the detection of influenza virus resistant strains.The fluorescent probe we synthesized in this study has the advantages of low cost,non-toxicity,convenient operation and strong signal.This makes the fluorescent probe a prospective method for detecting influenza viruses.
