Inflammatory bowel disease(IBD)is a chronic intestinal disease caused by a variety of unclear causes,including Crohn's disease(CD)and ulcerative colitis(UC).The incidence of IBD continues to rise,and the age of onset is getting younger,which may even increase the risk of patients suffering from colon cancer(CRC).The quality of life of IBD patients declines,making them more prone to depression and anxiety.Traditional treatment methods mainly treat inflammation and regulate immune disorders,but long-term large-dose application of immunosuppressants will increase the possibility of cancer and infection in patients.Although the exact pathogenesis of IBD has not been clarified,it is currently believed to be closely related to intestinal flora.Diet affects the intestinal flora and interferes with IBD.The prebiotics in the grain can be used as a growth promoter for beneficial bacteria in the intestine.The beneficial bacteria will inhibit inflammation to a certain extent.Studies have found that millet(Setaria italica(L.)Beauv.)contains a lot of prebiotics,and the content of minerals and polyphenol antioxidants in millet bran is high.And germination and fermentation of millet will not only improve protein digestibility,promote nutrient absorption in millet,but also reduce anti-nutritional factors such as phytic acid in bran.This study introduces the preparation of six different pre-treated grain flours for animal experiments.The millet is made into polished millet flour(M-001)?fermented flour of Polished millet(FM-002)?millet flour(BM-004)?fermented flour of Millet(BFM-005)?germinating millet flour(GBM-006)?fermented flour of Germinating millet(GBFM-007).The national standard determination method determined the nutritional components of six kinds of cereal flour.The nutrient components of the six kinds of grain flour are calculated and balanced,and added to the feed of mice.The feed code and the grouping code of the mice are the same as the grain flour code.The group fed with six kinds of grain flour supplemented feed was the experimental group,and the group fed with standard animal feed(AIN-93M)and refined wheat flour(RW-003)feed was the control group.A mouse model of colitis induced by dextran sodium sulfate(DSS)was constructed.DSS was induced for 5 days,during which the diet,drinking water,body weight and stool status of the mice were recorded,and stool samples were taken.After the experiment,the mice were anesthetized with isoflurane and sacrificed.Samples of the colon and other samples were taken to explore the effects of different pre-treated grain powders on intestinal inflammation in mice.The results showed that the mice were successfully modeled,diet and drinking water and body weight were gradually reduced due to colitis disease,and bloody stools and rectal bleeding symptoms appeared;the score of DAI value in the control group was significantly higher than that of the experimental group(Duncan test P ? 0.05),The lower one is M-001(5.5 ± 2.67),GBM-006(6 ± 2.44,)GBFM-007(6.4 ± 1.65)three groups,no significant difference,the lowest group is BFM-005 group,the value is(5.1 ± 2.81),the difference between these four groups was not significant;those with lower colon length were M-001,BM-004,BFM-005,GBM-006,GBFM-007,and there was no significant difference in colon length.Pathological sections of the mouse colon showed that the epithelial structures of the two control groups,AIN-93 M and RW-003,had been completely destroyed,and the villi crypts and gland structures disappeared.BFM-005,GBFM-006,GBFM-007 groups with lower tissue damage scores,the scores of these three groups were 3.6 ± 1.14?4.6 ± 1.52?3.4 ± 1.14 no significant difference.High-throughput sequencing of intestinal flora in mice,16 srDNA amplification sequencing analysis of microbial population composition and diversity,the results showed that the intestinal flora community diversity control group RW-003 is lower,GBFM-007 group High biodiversity,the most obvious inhibitory effect on conditionally pathogenic bacteria and the promotion of probiotic reproduction,which can balance intestinal flora and improve inflammation.This study demonstrates that developing a sound diet strategy is a viable potential approach to prevent and improve IBD disease.Millet germination and fermented food are more beneficial to intestinal health.
